DNA电击对草莓花粉活力的影响
Effects of DNA shock on the viability of strawberry pollen
【摘要】 以‘申琪’草莓花粉为材料,结合不同电脉冲形态、电场强度和电击次数进行处理,然后在花粉萌发培养基上28℃黑暗孵育,于3、6、18、24 h时用显微镜观察花粉萌发情况,探索草莓花粉电击适宜参数。结果表明:80μs方波脉冲在4.5—6.0 kV∕cm强度电击1次,可逆抑制花粉萌发,抑制程度与电场强度呈正相关;电击后3 h花粉萌发率为CK1的36%—70%,24 h时为CK1的60%—85%。方波4.5—5.0 kV∕cm强度电击2次产生部分可逆抑制,电击24 h后花粉萌发率为CK1的20%—50%;5.5—6.0 kV∕cm方波脉冲电击2次和4.5—6.0 kV∕cm电击3次产生不可逆抑制,电击24 h后花粉萌发率低于CK1的10%。用2.75—6.0 kV∕cm指数衰减波3.5 ms脉冲电击后,花粉活力快速下降,电击18 h后花粉萌发率在1%以下。综上,强度4.5—5.0 kV∕cm方波脉冲80μs电击2次为草莓花粉适宜电击参数。
【Abstract】 Taking ’Shenqi ’ strawberry pollens as the materials,combined with different electrical pulse forms,electric intensity and electric shock times,the pollens were incubated in the dark at 28 ℃ on the pollen germination medium.The pollen germination was observed and analyzed under a microscope at 3,6,18,and 24 h to explore the appropriate parameters for electric shock treatment of strawberry pollen.The results showed that an80 μs square wave pulse with 4.5—6.0 kV/cm electric intensity once reversibly inhibited pollen germination,and the inhibition degree was positively correlated with the electric intensity.The pollen germination rate after 3 h of electric shock was 36%—70% of CK1,and after 24 h it was 60%—85% of CK1.Partial reversible inhibition was generated by two times of electric shocks with a square wave intensity of 4.5—5.0 kV/cm,and the pollen germination rate was 20%—50% of CK1 after 24 h of electric shock.A square wave pulse of 5.5—6.0 kV/cm electric shock twice or 4.5—6.0 kV/cm electric shock 3 times resulted in irreversible inhibition,and the pollen germination rate was lower than 10% of CK1 after 24 h of treatment.After electric shocks with 2.75—6.0 kV/cm exponential decay wave for 3.5 ms,the pollen vitality rapidly decreased,and the pollen germination rate was below 1% after 18 h of electric shocks.In summary,a square wave pulse with an intensity of 4.5—5.0 kV/cm and an electric shock of 80 μs twice was the appropriate electric shock parameter for strawberry pollen.
