低强度聚焦超声联合顺铂对宫颈癌裸鼠皮下移植瘤抑制及增敏作用的实验研究

【摘要】 目的:探讨低强度聚焦超声(low-intensity focused ultrasound, LIFU)及LIFU联合顺铂对宫颈癌裸鼠皮下移植瘤的抑制及增敏作用。方法:将人宫颈癌HeLa细胞株接种于BALB/c雌性裸鼠,建立宫颈癌裸鼠皮下移植瘤模型,随机将造模成功的裸鼠分为空白组、顺铂组、LIFU组、LIFU联合顺铂组(联合组),经相应处理后,观察裸鼠一般状况及肿瘤生长情况,用HE染色观察肿瘤细胞病理组织学及形态学改变,TUNEL法检测不同治疗组的宫颈癌细胞凋亡情况。结果:LIFU组裸鼠的一般情况较好,而顺铂组裸鼠一般情况最差;在肿瘤组织HE染色中发现,LIFU组、顺铂组及联合组中均可见坏死区域,顺铂组坏死区域较LIFU组明显增多,联合组中坏死区域面积最大;与空白组相比,LIFU组、顺铂组、联合组抑瘤率分别为6.13%(P<0.05)、73.67%(P<0.01)、79.18%(P<0.01),差异均具有统计学意义。TUNEL法检测发现,LIFU组、顺铂组、联合组各组凋亡指数依次增加(P<0.05、P<0.01、P<0.01),联合组细胞凋亡情况最明显。结论:LIFU是一种较为安全的治疗方法,LIFU能抑制宫颈癌皮下移植瘤瘤体生长,LIFU与顺铂联用时,能有效提高顺铂的药物敏感性,增强对宫颈癌细胞的抑制作用,联合治疗对宫颈癌裸鼠皮下移植瘤组织的生长抑制作用明显优于单独使用顺铂组。

【Abstract】 Objective: To investigate the inhibitory and sensitizing effects of low-intensity focused ultrasound(LIFU) alone and LIFU combined with cisplatin on subcutaneously transplanted cervical cancer in nude mice. Methods: Human cervical cancer HeLa cells were inoculated into female BALB/c nude mice to establish a subcutaneous transplanted tumor model of cervical cancer. The successfully modeled mice were randomly assigned to the control group, the cisplatin group, the LIFU group, and the LIFU combined with cisplatin group(the combination group). After being treated, the general condition and tumor growth of the nude mice were observed. The pathological tissue and morphological changes in tumor cells were observed by HE staining, and the apoptosis of cervical cancer cells in different groups was detected by terminal deoxynucleotidyl transferase dUTP nick-end labeling(TUNEL) assay. Results: The general condition of the nude mice in the LIFU group was good, while that in the cisplatin group was the worst. The HE staining results revealed that necrotic areas were found in the LIFU group, the cisplatin group and the combination group, with a significant increase in the necrotic area in the cisplatin group compared to the LIFU group, and that the largest necrotic area was observed in the combination group. The tumor growth inhibition rates in the LIFU group, the cisplatin group, and the combination group were 6.13%(P<0.05), 73.67%(P<0.01) and 79.18%(P<0.01) which were significantly different from that in the control group. The TUNEL assay revealed that the apoptosis indexes in the LIFU group, the cisplatin group and the combination group increased(P<0.05, P<0.01, P<0.01), and the combination group showed the most obvious cell apoptosis. Conclusion: LIFU is a relatively safe treatment that can inhibit the growth of subcutaneously transplanted cervical cancer. LIFU combined with cisplatin can effectively improve its sensitivity to cisplatin, and enhance its inhibitory effect on cervical cancer cells. The combination therapy has a significantly better inhibitory effect on the growth of subcutaneously transplanted cervical cancer in nude mice than the single use of cisplatin.
