不同抗凝剂浓度对大鼠凝血四项检测结果的影响
The Influence of Different Anticoagulant Concentrations on Hemostatic Parameters in Rat
【摘要】 目的 探讨不同抗凝剂浓度对大鼠凝血四项检测结果的影响。方法 10只SPF级SD成年雄鼠真空采血法采集后腔静脉血液,每只大鼠采集4管血,使得各管抗凝比(枸橼酸钠抗凝剂与全血体积比)分别为1∶9、1∶7、1∶5、1∶3。分离血浆,全自动血凝分析仪检测凝血四项指标。结果 随着抗凝剂浓度的增加,APTT逐渐延长;FIB逐渐减少;PT呈现轻微的上升趋势;TT呈现下降趋势。APTT以及FIB在1∶9和1∶7两个浓度间差异无统计学意义(P>0.05),当抗凝剂浓度增加至1∶5和1∶3时,APTT和FIB与各浓度间的差异均具有统计学意义(P<0.05);PT在1∶7和1∶5两个浓度间差异无统计学意义(P>0.05),当浓度为1∶9和1∶3时,PT与其他各浓度间的差异均具有统计学意义(P<0.05);TT在每个浓度下与各浓度间差异均具有统计学意义(P<0.05)。结论 不同抗凝剂浓度对大鼠凝血功能有显著影响。为了在大鼠实验中获得准确且稳定的检测结果,建议将抗凝剂浓度控制在1∶7~1∶9。
【Abstract】 Objective To investigate the effects of different anticoagulant concentrations on the hemostatic results in rats.Methods Ten adult male SPF-grade SD rats were subjected to vacuum blood collection from the posterior vena cava.Four blood samples were collected from each rat, achieving anticoagulant ratios(sodium citrate anticoagulant to whole blood volume ratio) of 1∶9,1∶7,1∶5,and 1∶3 in each tube.Plasma was separated, and the hemostatic parameters were performed using a fully automated coagulation analyzer.Results As the concentration of anticoagulant increased, APTT gradually prolonged, FIB decreased, PT showed a slight upward trend, and TT exhibited a downward trend.APTT and FIB showed no statistically significant difference between the concentrations of 1∶9 and 1∶7(P>0.05).However, as the anticoagulant concentration increased to 1∶5 and 1∶3,both APTT and FIB exhibited statistically significant differences between each concentration(P<0.05).PT displayed no statistically significant difference between the concentrations of 1∶7 and 1∶5(P>0.05).When the concentration was 1∶9 and 1∶3,PT showed statistically significant differences between each concentration(P<0.05).TT demonstrated statistically significant differences between each concentration(P<0.05).Conclusion Different concentrations of anticoagulants have significant effects on the coagulation function of rats.To obtain accurate and stable test results in rat experiments, it is recommended to maintain the anticoagulant concentration between 1∶7 and 1∶9.
