白藜芦醇对鼠伤寒沙门氏菌诱导型IPEC-J2细胞损伤的缓解作用
Alleviating effect of resveratrol on Salmonella typhimurium-induced damage in IPEC-J2 cell
【摘要】 【目的】研究白藜芦醇(resveratrol, RES)对鼠伤寒沙门氏菌(Salmonella typhimurium)诱导猪小肠上皮细胞(intestinal porcine epithelial cells, IPEC-J2)损伤的缓解作用,为促进动物健康养殖提供新思路。【方法】用鼠伤寒沙门氏菌建立IPEC-J2损伤模型,测定细胞上清液一氧化氮(NO)浓度、肿瘤坏死因子-α(TNF-α)质量浓度、白介素-6(IL-6)质量浓度、白介素-4(IL-4)质量浓度、乳酸脱氢酶(lactate dehydrogenase, LDH)水平、总超氧化物歧化酶(total superoxide dismutase, T-SOD)活力、还原型谷胱甘肽(glutathione, GSH)质量浓度和丙二醛(malondialdehyde, MDA)浓度,分析RES对IPEC-J2损伤模型的缓解作用。【结果】RES预处理IPEC-J2细胞的最佳质量浓度为7.8 mg·L-1,且能有效缓解鼠伤寒沙门氏菌诱导IPEC-J2细胞产生的炎症损伤,显著降低促炎因子(TNF-α和IL-6)的质量浓度,提高抑炎因子(IL-4)的表达,进而有效减缓肠道炎症反应。同时,RES可显著提高T-SOD活力和GSH质量浓度,显著降低MDA浓度,缓解鼠伤寒沙门氏菌诱发的氧化损伤,达到维持机体肠道氧化还原平衡的稳态。【结论】RES对IPEC-J2损伤具有保护作用,可用于预防鼠伤寒沙门氏菌感染。
【Abstract】 【Objective】To investigate the protective effects of resveratrol(RES) against oxidative and inflammatory damage induced by Salmonella typhimurium in intestinal porcine epithelial(IPEC-J2) cells and provide insights to enhance animal health in breeding practices.【Method】An IPEC-J2 cell injury model was established using S.typhimurium, and key oxidative stress and inflammatory indicators were measured in the cell supernatant, including nitric oxide(NO) content, tumor necrosis factor-α(TNF-α), interleukin-6(IL-6), interleukin-4(IL-4) secretion, lactate dehydrogenase(LDH) level, total superoxide dismutase(T-SOD), reduced glutathione(GSH) and malondialdehyde(MDA), to analyze the mitigating effects of RES on IPEC-J2 cell injury model. 【Result】Pretreatment of IPEC-J2 cells with RES at an optimal mass concentration of 7.8 mg·L-1 could significantly alleviate the inflammatory injury induced by S.typhimurium, notably reducing the contents of pro-inflammatory factors(TNF-α and IL-6), and increasing the expression of inhibitory factors(IL-4), thus effectively slowing down the intestinal inflammatory response. Meanwhile, RES could significantly increase the activities of T-SOD and GSH enzymes, significantly reduce the expression level of MDA, and alleviate the oxidative damage induced by S.typhimurium, thereby contributing to the maintenance of intestinal redox homeostasis of the organism. 【Conclusion】RES has a protective effect on IPEC-J2 cell damage and holds potential for preventing S.typhimurium infection in animal models.
