富硒海鲜菇菌丝体固体发酵的工艺优化
Optimization of solid fermentation process for enriching selenium on mycelium of Hypsizygus marmoreus
【摘要】 为提高海鲜菇菌粉中多糖及其中硒的含量,对海鲜菇菌丝体富硒固体发酵进行了研究和工艺条件的优化。在确定海鲜菇菌丝体耐受亚硒酸钠浓度范围的基础上,以多糖得率、多糖硒含量和长势评分为指标,考察了亚硒酸钠浓度、富硒菌液添加量和接种量对海鲜菇菌丝体富硒固体发酵的影响,最后采用响应面法优化得到固体发酵的条件。结果表明,海鲜菇菌丝体在0~100 mg·L-1亚硒酸钠浓度范围内可正常生长;最佳发酵条件为亚硒酸钠浓度29 mg·L-1、富硒菌液添加量45%、接种量15%,在此发酵条件下,菌丝体中多糖得率和多糖硒含量分别为(22.90±1.62)%和(316.37±1.85) mg·kg-1。该发酵方法切实可行,富硒效果显著,可为菌丝体富硒发酵提供一定的参考价值。
【Abstract】 In order to increase the content of polysaccharide and selenium in Hypsizygus marmoreus powder, the solid fermentation process for enriching selenium on the mycelium of Hypsizygus marmoreus was applied for optimization for the technological conditions. After determining the range of sodium selenite tolerance in the mycelia, the effects of sodium selenite concentration, added amount of selenium enriched bacterial solution and inoculation dosage of Selenium-rich strains on the fermentation was investigated by evaluating polysaccharide yield, selenium content in polysaccharide and growth score. Finally,the response surface method was used to optimize the solid fermentation conditions. The results showed that mycelia of Hypsizygus marmoreus could grow normally in the concentration range of 0~100 mg·L-1 sodium selenite. The optimal fermentation conditions were 29 mg·L-1 of sodium selenite concentration, 45% of addition amount of selenium-rich bacterial solution and 15% of inoculation amount. The yield of polysaccharides and the selenium content of polysaccharides in mycelium were(22.90±1.62)%and(316.37±1.85) mg·kg-1, respectively Therefore, the solid fermentation is feasible to enriching selenium on mycelium and a potential method for selenium-rich mycelium.
