HPLC法同时测定星夏止痛贴膏中3种双酯型生物碱含量

【摘要】 目的:建立星夏止痛贴膏中生川乌、生草乌所含双酯型生物碱成分含量的HPLC同时测定方法。方法:采用Phenomenex Luna Omega PS C18(4.6 mm×250 mm, 5μm)色谱柱分离,乙腈-四氢呋喃(25∶15)-0.1 moL/L醋酸铵为流动相梯度洗脱,检测波长:235 nm;柱温:25℃;流速:1.0 mL/min,外标法定量。结果:3种双酯型生物碱成分分离度良好,分别在14.151~113.207μg/mL、13.392~107.136μg/mL、14.196~113.564μg/mL浓度范围内与峰面积呈良好的线性关系,平均回收率分别为94.03%、97.92%、94.45%,RSD分别为5.51%、7.09%、6.94%。结论:该方法简便、快速、准确,可同时对星夏止痛贴膏中的乌头碱、次乌头碱、新乌头碱3双酯型生物碱进行分析,能有效地消除杂质干扰,为星夏止痛贴膏的质量评价提供实验依据。

【Abstract】 Objective A high performance liquid chromatography(HPLC)was established for the simultaneous determination of 3 diester diterpenoid alkaloids, i.e. mesaconitine, aconitine and hypaconitine in Xingxia Zhitong plaster. Methods The 3 alkaloids were separated on Phenomenex Luna Omega PS C18(4.6 mm×250 mm, 5 μm)column with acetonitrile-tetrahydrofuran(25∶15)-ammonium acetate as the mobile phase by gradient elution at a flow rate of 1.0 mL·min-1. The detective wavelength was235 nm, the column temperature was 25 ℃,and finally quantified by external standard calibration method.Results The 3 alkaloids showed a good linearity relationship between 14.151-113.207 μg/mL、13.392-107.136 μg/mL、14.196-113.564 μg/mL,and the average recoveries were 94.03%(RSD=5.51%)、97.92%(RSD=7.09%)、94.45%(RSD=6.94%), respectively.Conclusion The method is rapid, simple and accurate, it can simultaneously analyze the contents of 3 alkaloids in Xingxia Zhitong plaster and eliminate the impurity interference effectively. This method and test data also provided reference for the quality evaluation of Xingxia Zhitong plaster.
