牡丹拮抗内生细菌的筛选及发酵条件优化研究

【摘要】 以牡丹根部组织为材料,对其根部内生细菌进行分离纯化,筛选出能够对金黄色葡萄球菌产生拮抗作用的菌株,并根据常规分类学结合16S rRNA对筛选出的菌株进行鉴定.通过单因素试验和正交试验确定了培养基成分的最佳组合,并对发酵条件进行了优化.结果表明:菌株mudan-6对金黄色葡萄球菌的拮抗作用最好,抑菌圈直径为(20±0.3)mm,经测序鉴定该菌株为产气克雷伯氏菌(Klebsiella aerogenes);最佳培养基组合为0.5%蔗糖,1.5%酵母浸粉,1%碳酸钙;最佳发酵条件为接种量5%,发酵温度37℃, pH值9.0,装液量100 mL,转速180 r/min,培养时间12 h.

【Abstract】 The root tissue of paeonia suffruticosa was used as the experimental material, and the endophytic bacteria in the root were isolated and purified to screen out the antagonistic strains against Staphylococcus aureus, and the screened strains were identified according to conventional taxonomy combined with 16S rRNA. The optimal combination of medium components was determined by single factor test and orthogonal test, and the fermentation conditions were also optimized. The results showed that the strain mudan-6 had the best antagonistic effect against Staphylococcus aureus, and the diameter of inhibition zone was(20±0.3)mm; the strain was identified as Klebsiella aerogenes by sequencing; the optimal medium was 0.5% sucrose, 1.5% yeast extract and 1% calcium carbonate; the optimum fermentation conditions were as follows: inoculum size 5%, fermentation temperature 37 ℃, pH value 9.0, liquid volume 100 mL, rotation speed 180 r/min, and culture time 12 h.
