超活化血小板裂解液对大鼠牙周骨改建影响的实验研究
An experimental study on the effect of super-activated platelet lysate on periodontal bone remodeling in rats
【摘要】 目的 评估超活化血小板裂解液(super-activated platelet lysate, sPL)与明胶海绵联合应用对大鼠牙周骨缺损骨重建的影响,以期为临床牙周骨组织再生治疗提供实验参考。方法 取16只8周龄雄性SD大鼠建立双侧牙周骨缺损模型共32个。随机分为4组:明胶海绵+sPL组、明胶海绵组、sPL组、对照组。术后第4周和第8周分别处死相应大鼠并取上颌骨标本。微计算机断层扫描技术(micro computed tomography, Micro-CT)评估牙周骨成骨情况。HE染色观察牙周骨组织形态变化。结果 在第4周,骨矿化密度(bone mineral density, BMD)、骨体积分数(bone volume fraction, BV/TV)、骨小梁数量(trabecular number, Tb.N)、骨小梁厚度(trabecular thickness, Tb.Th)测量指标,各组均无统计学意义(P>0.05)。第8周明胶海绵+sPL组BMD、BV/TV、Tb.N显著高于对照组,差异有统计学意义(P<0.05);Tb.N测量值明胶海绵+sPL组较sPL组高,差异有统计学意义(P<0.05)。Tb.Th测量值差异无统计学意义(P>0.05)。组织学观察结果显示,相比其他组,明胶海绵+sPL组的新骨形成最多,牙周骨缺损基本愈合。结论 sPL与明胶海绵联合应用能促进大鼠牙周骨缺损的骨改建,促进牙周骨组织愈合。
【Abstract】 Objective To evaluate the effect of combined application of super-activated platelet lysate(sPL) and gelatin sponge on bone reconstruction of periodontal bone defects in rats, in order to provide experimental reference for clinical periodontal bone tissue regeneration treatment. Methods A total of 32 bilateral periodontal bone defect models were established in 16 8-week-old male SD rats. They were randomly divided into 4 groups, namely gelatin sponge+sPL group, gelatin sponge group, sPL group, and control group. The corresponding rats were sacrificed at the 4th and 8th weeks after surgery, and maxillary bone specimens were collected. Micro-CT was evaluated the osteogenesis of periodontal bone. HE staining was used to observe the morphological changes of periodontal bone tissues. Results In the 4th week, there was no statistical significance in the measurement indicators of BMD, BV/TV, Tb.N, and Tb.Th in each group(P>0.05). The 8th week showed that the BMD, BV/TV, and Tb.N of the gelfoam+sPL group were significantly higher than those of the control group, and the difference was statistically significant(P<0.05); the Tb.N measurement value of the gelfoam+sPL group was higher than that of the sPL group, and the difference was statistically significant(P<0.05). The difference in Tb.Th measurement values was not statistically significant(P>0.05). Histological observation showed that compared with other groups, the gelatin sponge+sPL group had the most new bone formation, and the periodontal bone defects were basically healed. Conclusion The combined application of sPL and gelatin sponge could promote bone reconstruction of periodontal bone defects in rats and promote periodontal bone tissue healing.
