不同笋用竹内生真菌种类组成及其多样性

【摘要】 【目的】为了解笋用竹内生真菌多样性以及其在不同组织器官中的分布特征。【方法】分别选取雷竹、厚竹和高节竹的健康根、茎、叶植株样品,经表面消毒后接种到PDA培养基,对菌株进行分离纯化并鉴定。【结果】从3种笋用竹中分离出191株内生真菌,优势菌属为Apiospora属、Nigrospora属、Arthrinium属、Shiraia属。不同组织器官中内生真菌的数量和种类存在差异。叶片内生真菌为147株,多样性指数高于茎和根。不同笋用竹中,厚竹75株,菌株定殖率为41.48%;雷竹最少(54株),菌株定殖率为35.56%。不同笋用竹内生真菌多样性指数和均匀度为雷竹>高节竹>厚竹,高节竹和雷竹内生真菌多样性指数均表现为叶>根>茎,而厚竹内生真菌多样性指数表现为叶部>茎部>根部。相似性分析表明,高节竹叶和厚竹叶内生真菌相似性较高。【结论】笋用竹内生真菌具有丰富的多样性,不同笋用竹及其组织器官间内生真菌群落多样性存在差异性,并具有宿主专一性和组织偏好性。

【Abstract】 [Objective] This study was conducted in order to understand the diversity of endophytic fungi in bamboo species for bamboo shoots and their distribution characteristics in different tissue organs. [Method] Root,stem and leaf samples from healthy plants of Lei bamboo forests, thick bamboo forests and tall bamboo forests were surface sterilized and then inoculated into PDA medium to isolate and purify and identify the endophytic fungi. [Result] 191 endophytic fungi were isolated from three species of bamboo. The dominant fungal genera of bamboo were Apiospora spp, Nigrospora spp, Arthrinium spp, and Shiraia spp. and the number and species of endophytic fungi and their diversity in different tissues and organs differed greatly. The leaves had the most endophytic fungi(147 strains), with the higher diversity index than that of stems and roots. Among different bamboo species, thick bamboo had the most(75 strains), with a strain colonization rate of 41.48%; Lei bamboo had the least(54 strains), with a strain colonization rate of 35.56%. The diversity index and uniformity of endophytic fungi in different bamboo species were: Lei bamboo>high knotted bamboo>thick bamboo, and both high knotted bamboo and Lei bamboo showed a higher diversity index of endophytic fungi for leaves>roots >stems, while the endophytic fungal diversity index of thick bamboo was expressed in leaves >stems>roots. The similarity analysis showed that the endophytic fungi were more similar in high nodal bamboo leaves and thick bamboo leaves. [Conclusion] Endophytic fungi in bamboo species are rich in diversity, and there is variability in endophytic fungal community diversity among different bamboo species, their tissues and organs, with host specificity and tissue preference.
