区分肺腺癌和肺鳞状细胞癌的潜在基因分析
Potential Gene Analyze for Distinguishing Lung Adenocarcinoma from Lung Squamous Cell Carcinoma
【摘要】 目的 对非小细胞肺癌(NSCLC)潜在基因进行分析,寻找区分肺腺癌(LUAD)和肺鳞状细胞癌(LUSC)的相关基因,探索NSCLC诊断与治疗的更有效方法。方法 利用3个基因表达综合(GEO)数据库(GSE4882、GSE7339和GSE40275)分析LUAD和LUSC组织样本中的差异表达基因(DEGs),对DEGs进行基因本体(GO)富集分析并构建蛋白质-蛋白质相互作用(PPI)网络来寻找关键枢纽基因。最后在临床样本中验证关键枢纽基因的差异表达。结果 在GSE4882、GSE7339和GSE40275数据集中发现22个LUAD和LUSC的DEGs,通过构建PPI网络发现了6个关键枢纽基因(KRT18、RAN、NME1、NME2、MIF和CFB),进一步在肿瘤基因组图谱(TCGA)数据库中分析枢纽基因表达与生存之间的关系,发现KRT18可能是区分LUAD和LUSC的潜在基因,同时构建了KRT18基因突变与LUAD和LUSC患者预后的风险预测模型。最后采用新乡医学院第一附属医院临床组织样本对KRT18表达进行验证。结论 KRT18可能是区分LUAD和LUSC潜在基因。
【Abstract】 Objective To analyze the potential genes of non-small cell lung cancer(NSCLC), and to identify relevant genes that distinguish lung adenocarcinoma(LUAD) from lung squamous cell carcinoma(LUSC), and explore more effective methods for the diagnosis and treatment of NSCLC.Methods The differentially expressed genes(DEGs) in LUAD and LUSC tissue samples was analyzed by three Gene Expression Omnibus(GEO) databases(GSE4882, GSE7339 and GSE40275). Gene Ontology(GO) enrichment analysis was performed on DEGs and protein-protein interaction(PPI) network was constructed to identify key hub genes. Finally, the differential expression of key hub genes was verified in clinical samples.Results Twenty-two DEGs of LUAD and LUSC were found in GSE4882, GSE7339 and GSE40275 datasets. Six key hub genes(KRT18, RAN, NME1, NME2, MIF and CFB) were discovered by constructing a PPI network. The relationship between hub gene expression and survival in The Cancer Genome Atlas(TCGA) database was analyzed, and revealed that KRT18 may be a potential gene for distinguishing LUAD from LUSC. Meanwhile, a risk prediction model for KRT18 gene mutation and prognosis of LUAD and LUSC patients was constructed. Finally, clinical tissue samples from the First Affiliated Hospital of Xinxiang Medical College were used to verify the KRT18 expression.Conclusion KRT18 may be a potential gene marker to distinguish LUAD from LUSC.
