发酵乳杆菌GBJ荚膜多糖结构、抗氧化及免疫活性研究
Study on Structure,Antioxidant and Immune Activity of Capsular Polysaccharides from Lactobacillus Fermentum GBJ
【摘要】 采用酶法提取发酵乳杆菌GBJ荚膜多糖(Capsular polysaccharides,CPS),并对其初级结构、体外抗氧化及免疫活性进行研究。结果表明:Lactobacillus fermentumGBJ CPS的产量为124.37 mg·L-1,其中性糖含量为92.54%,蛋白质含量为3.06%,硫酸基含量为1.04%,且不含糖醛酸;单糖组成分析显示CPS主要由摩尔比为3.64:1.00的半乳糖和葡萄糖组成;红外分析结果表明CPS具有多糖的特征吸收峰。此外,CPS在体外表现出较高的DPPH、ABTS+和羟基自由基清除能力。同时,合适浓度的CPS还可显著提高RAW264.7细胞的中性红吞噬能力和细胞因子(IL-1β、IL-6和IL-10)分泌水平(P<0.05),表明其具有良好的免疫调节作用。研究可以为乳酸菌CPS未来的开发和应用提供理论依据。
【Abstract】 The capsular polysaccharides(CPS) ofLac tobac illus fermentumGBJ were extracted with enzymes and the primary structure,antioxidant activity in vitro and immune activities of the CPS were investigated.The results showed that the CPS production ofL.ferme ntumGBJ was 124.37 mg·L-1,with neutral sugar content of 92.54%,protein content of 3.06%,sulfate group content of1.04%,and no uronic acid.The monosaccharide composition showed that CPS was mainly composed of galactose and glucose with molar ratio of 3.64 :1.00.Infrared analysis showed that CPS had characteristic absorption peak of polysaccharide.In addition,CPS showed higher DPPH,ABTS+and hydroxyl radical scavenging abilities in vitro.At the same time,appropriate concentration of CPS could significantly improve the neutral red phagocytosis and the secretion levels of cytokines(IL-1β,IL-6 and IL-10)in RAW264.7cells(P<0.05),which indicated that CPS had a good immunomodulatory effect.It could provide theoretical basis for the future development and application of lactic acid bacteria CPS.
