黄瓜未授粉子房离体培养体系影响因素
The Study on Influencing Factors of in Vitro Culture System of Cucumber Unpollinated Ovary
【摘要】 为筛选出适合黄瓜多种基因型的胚诱导体系,以‘翡翠’‘津优35’‘瑞光2号’‘京丰268’等4种不同基因型的黄瓜为试材,对黄瓜未授粉子房离体培养的影响因素进行研究。结果表明,在不同组合处理中,4种基因型的试材均表现为花前2 d的黄瓜未授粉子房采用MS培养基中添加0.07 mg·L-1噻苯隆(TDZ),35℃热激处理2 d的胚发生率最高,分别为60.8%,20.8%,43.3%,65.0%。不同基因型间胚发生率差异显著,其中,‘京丰268’胚发生率最高,为18.04%,‘瑞光2号’胚发生率最低,为5.23%。35℃热激处理2 d的效果最好,开花前2 d的未授粉子房胚发生率最高。‘翡翠’以0.09 mg·L-1 TDZ处理时胚发生率最高,‘瑞光2号’‘津优35’‘京丰268’以0.07 mg·L-1 TDZ处理时胚发生率最高。
【Abstract】 Taking cucumber species, ‘Fei Cui’ ‘Jin You 35’ ‘Rui Guang 2’ and ‘Jing Feng 268’ as test materials, influencing factors of in vitro culture of cucumber unpollinated ovary were studied to screen out the embryo induction system suitable for various genotypes of cucumber. The results showed that in different combinations of the four genotypes, the embryo incidence of unpollinated ovary of cucumber was the highest in 35 ℃ heat shock treatment for 2 days with 0.07 mg·L-1 TDZ in MS medium 2 days before flowering, which was 60.8%, 20.8%, 43.3% and 65.0%, respectively. There was a significant difference in the incidence of embryos among different genotypes, the incidence of ‘Jing Feng 268’ was the highest with 18.04%, and that of ‘Rui Guang 2’ was the lowest, with 5.23%. The effect of heat shock treatment at 35 ℃ for 2 days was the best, and the incidence of unpollinated ovary embryo was the highest 2 days before flowering. The embryo incidence rate of ‘Fei Cui’ was the highest when treated with 0.09 mg·L-1 TDZ, and that of ‘Rui Guang 2’ ‘Jin You 35’ and ‘Jing Feng 268’ was the highest when treated with 0.07 mg·L-1 TDZ.
