老鼠簕来源内生真菌Diaporthe pseudomagiferae的聚酮类化合物分离鉴定及体外抗氧化、抗炎活性研究
Isolation and identification of polyketides from the endophytic fungus Diaporthe pseudomagiferae derived from Acanthus ilicifolius L. and studies on their anti-oxidant and anti-inflammatory activities in vitro
【摘要】 目的:研究老鼠簕来源的内生真菌Diaporthe pseudomagiferae中聚酮类化合物分离纯化及其抗氧化、抗炎活性。方法:利用正相硅胶柱色谱、Sephadex LH-20凝胶柱色谱、高效液相色谱等色谱技术进行分离纯化,结合高分辨质谱、核磁共振波谱技术等综合谱学技术对化合物进行结构解析,对部分化合物进行抗炎活性评价和抗氧化活性筛选。结果:从Diaporthe pseudomagiferae大米发酵产物中分离鉴定得到7个聚酮类化合物,分别为:[(8-methyl-9-oxabicyclo [4.2.1] nona-2,4-dien-7-yl)-(5-oxo-2H-furan-2-yl)methyl] acetate(1)、2,3-Dihydromycoepoxydiene(2)、Dothiorelone A(3)、Dothiorelone B(4)、Cytosporone B(5)、5,6-dihydroxy-2,3,6-trimethylcyclohex-2-enone(6)、3-(4-Methylphenoxy)propanoic acid(7)。1,1-二苯基-2-三硝基苯肼(DPPH)自由基清除实验结果显示,化合物1~5均未表现出抗氧化活性。酶联免疫吸附试验(ELISA)结果显示,化合物1~5对脂多糖(LPS)诱导的RAW264.7细胞释放的炎症因子白细胞介素-6(IL-6)有一定的抑制作用。结论:化合物1、3、4和6首次从老鼠簕内生真菌Diaporthe pseudomagiferae中分离得到,化合物1~5具有一定的抗炎活性,为进一步探索相关活性物质的抗炎机制奠定基础。
【Abstract】 Objective: To investigate the isolation and purification of the polyketide compounds and their anti-oxidant and anti-inflammatory activities in the endophytic fungus Diaporthe pseudomagiferae from Acanthus ilicifolius L. Methods: The compounds were separated and purified by column chromatography on silica gel, Sephadex LH-20, and high-performance liquid chromatography, and the structures of compounds were analyzed by combining with comprehensive spectral techniques such as high resolution mass spectra(MS) and nuclear magnetic resonance(NMR) spectroscopy. The anti-inflammatory and anti-oxidant activities of some pure compounds were evaluated. Results: A total of seven polyketides were isolated from rice fermentation products of Diaporthe pseudomagiferae and identified as [(8-Methyl-9-oxabicyclo [4.2.1] nona-2,4-dien-7-yl)-(5-oxo-2H-furan-2-yl) methyl] acetate(1), 2,3-Dihydromycoepoxydiene(2), Dothiorelone A(3), Dothiorelone B(4), Cytosporone B(5), 5,6-dihydroxy-2,3,6-trimethylcyclohex-2-enone(6), 3-(4-Methylphenoxy) propanoic acid(7). The free radical scavenging experiment of 1, 1-diphenyl-2-trinitrophenylhydrazine(DPPH) showed that compounds 1-5 did not exhibit anti-oxidant activity. The results of enzymed-linked immunosorbent assay(ELISA) showed that compounds 1-5 had inhibitory effects on interleukin-6(IL-6) release from RAW264.7 cells induced by lipopolysaccharide(LPS). Conclusion: Compounds 1, 3, 4, and 6 have been isolated from the endophytic fungus Diaporthe pseudomagiferae from Acanthus ilicifolius L. for the first time. Compounds 1-5 exhibit certain anti-inflammatory activity,which lays the foundation for further exploring the anti-inflammatory mechanisms of relevant active substances
