CDK14和FOLR1在结肠癌组织中的表达及与患者临床病理特征和预后的关系

【摘要】 目的 分析周期蛋白依赖性激酶14(CDK14)、叶酸结合蛋白1(FOLR1)在结肠癌组织中的表达及与患者临床病理特征和预后的关系。方法 选择2016年10月至2019年10月由洪湖市人民医院收治的177例结肠癌患者作为研究对象,收集其经手术切除的结肠癌组织及癌旁组织。采用实时荧光定量PCR法检测组织中CDK14、FOLR1的mRNA表达水平。采用免疫组织化学法检测组织中CDK14、FOLR1的蛋白表达情况。采用Pearson法分析结肠癌组织中CDK14、FOLR1表达的相关性。采用Kaplan-Meier生存曲线分析结肠癌组织中CDK14、FOLR1表达与患者预后的关系(采用log-rank检验)。采用COX回归分析探讨结肠癌患者预后的危险因素。结果 结肠癌组织中CDK14、FOLR1的m RNA表达水平及其蛋白表达阳性率均显著高于癌旁组织(P均<0.05)。Pearson相关性分析结果显示,结肠癌组织中CDK14与FOLR1表达呈正相关(P<0.05)。结肠癌组织中CDK14、FOLR1表达与患者的TNM分期、分化程度和浸润深度均有相关性(P均<0.05)。Kaplan-Meier生存曲线分析结果显示,结肠癌组织中CDK14、FOLR1阳性表达的患者的3年累积生存率分别显著低于CDK14和FOLR1阴性表达的患者(P均<0.05)。COX回归分析结果显示,结肠癌组织中CDK14、FOLR1阳性表达均是结肠癌患者预后的危险因素(P均<0.05)。结论 CDK14、FOLR1在结肠癌组织中表达水平均显著升高,并且两者与患者的TNM分期、分化程度、浸润深度和预后均密切相关,CDK14、FOLR1阳性表达均是结肠癌患者预后的危险因素。

【Abstract】 Objective This paper aims to analyze the expression of cyclin dependent kinase 14(CDK14) and folate binding protein 1(FOLR1) in colon cancer and their relationship with clinicopathological characteristics and prognosis of patients. Methods A hundred and seventy-seven colon cancer patients admitted to Honghu People’s Hospital from October 2016 to October 2019 were selected as the research subjects, and their colon cancer tissues and adjacent tissues were collected after surgical resection. Real time fluorescence quantitative PCR(qRT-PCR) was used to detect the mRNA expression levels of CDK14 and FOLR1 in tissues while the immunohistochemical method was used to detect the protein expression of CDK14 and FOLR1 in tissues. The Pearson method was used to analyze the correlation between CDK14 and FOLR1 expression in colon cancer tissues. The Kaplan Meier survival curve was used to analyze the relationship between CDK14 and FOLR1 expression in colon cancer tissues and patient prognosis(using Log-rank test). The COX regression analysis was used to explore the risk factors for prognosis in colon cancer patients. Results The m RNA expression levels and protein expression positivity rates of CDK14 and FOLR1 in colon cancer tissues are significantly higher than those in adjacent tissues(P<0.05). The Pearson correlation analysis results show a positive correlation between CDK14 and FOLR1 expression in colon cancer tissues(P<0.05). The expression of CDK14 and FOLR1 in colon cancer tissue is correlated with TNM staging, differentiation degree, and infiltration depth of patients(P<0.05). The Kaplan-Meier survival curve analysis results show that the 3-year cumulative survival rates of patients with CDK14 and FOLR1 positive expression in colon cancer tissue are significantly lower than those of patients with CDK14 and FOLR1 negative expression, respectively(P<0.05). The results of COX regression analysis show that the positive expression of CDK14 and FOLR1 in colon cancer tissues are risk factors for the prognosis of colon cancer patients(P<0.05). Conclusion The expression levels of CDK14 and FOLR1 are significantly increased in colon cancer tissues, and both are closely related to the TNM staging, differentiation degree, infiltration depth, and prognosis of patients, and the positive expression of CDK14 and FOLR1 are risk factors for the prognosis of colon cancer patients.
