原钙黏蛋白7的结构和功能及与肿瘤的关系

【摘要】 钙黏附蛋白是一类钙依赖性的黏附分子,是一种细胞跨膜糖蛋白,介导正常组织、肿瘤组织中细胞-细胞间的黏附作用,原钙黏蛋白(protocadherin, PCDH)是其最大的亚家族。PCDH广泛分布于中枢神经系统,根据基因结构分为非成簇原钙黏蛋白和成簇原钙黏蛋白。PCDH7属于非成簇PCDHs家族,在人类基因组中定位于4p15,主要在人类的脑和心脏中表达,具有与经典钙黏附蛋白相似的作用,介导细胞之间的识别和黏附。在多种肿瘤中均检测到不同程度PCDH7的异常表达,PCDH7可能与肿瘤的增殖、侵袭、转移和预后相关。本文对PCDH7的基因结构和功能与肿瘤的关系作一综述。

【Abstract】 Cadherins are a class of calcium-dependent adhesion molecules, which are transmembrane glycoproteins that mediate cell-cell adhesion in normal and tumor tissues. Protocadherins(PCDHs) represent the largest subfamily of cadherins. PCDHs are widely distributed in the central nervous system and can be classified into non-clustered protocadherins and clustered protocadherins based on their gene structure. PCDH7 belongs to the non-clustered PCDHs family and is located on chromosome 4p15 in the human genome. It is mainly expressed in the human brain and heart and exhibits similar functions to classical cadherins, mediating cell recognition and adhesion. Aberrant expression of PCDH7 has been detected to varying degrees in various tumors, and it may be associated with tumor proliferation, invasion, metastasis, and prognosis.
