木薯NHX基因家族鉴定及特征分析
Identification and Characteristics Analysis of NHX Gene Family in Manihot esculenta
【摘要】 【目的】鉴定木薯NHX基因家族成员并分析特征,为研究木薯耐盐新种质和挖掘木薯抗逆特性基因具有重要意义。【方法】基于全基因组水平上对木薯NHX基因家族进行鉴定,对其系统进化树、蛋白理化性质、保守结构、保守基序、共线性、蛋白互作等进行分析。【结果】在木薯中共鉴定到9条MeNHX基因(MeNHX1~MeNHX9),分为3个亚族,且不均匀地分布在7条染色体上,其中,染色体Chr03和Chr15中均包含2条MeNHX基因,染色体Chr01、Chr08、Chr09、Chr10和Chr17均只有1条MeNHX基因。MeNHX蛋白有520~1 157个氨基酸,其相对分子质量为58.09~128.21 kDa,理论等电点为5.63~9.11;为两性蛋白,不稳定系数为34.60~41.34;为疏水性蛋白,其GRAVY为0.094~0.595。9条MeNHX基因均含有典型的Na~+/H~+保守结构域,木薯NHX存在多个共线性区块,进化压力Ka/Ks值均小于1,主要受纯化选择作用;含有光响应、生物胁迫及非生物胁迫作用相关元件MeNHX基因同多条离子通道基因存在蛋白互作关系。【结论】MeNHX基因在木薯受到光、生物胁迫和非生物胁迫时参与生长发育过程,以适应其环境变化。
【Abstract】 【Objective】The NHX gene family members in Manihot esculenta were investigated and the characteristics were analyzed to provide a reference for the study of new M. esculenta germplasm with resistance to salt and the exploration of stress resistance genes in M. esculenta. 【Method】The NHX gene family of M. esculenta was identified based on the genome level, and its phylogenetic tree, protein physicochemical properties, conserved structure, conserved motifs, collinearity and protein interactions were analyzed.【Result】9 MeNHX genes(MeNHX1—MeNHX9) were identified from M. esculenta, which were divided into 3 subfamilies and unevenly distributed on 7 chromosomes. The chromosomes Chr03 and Chr15 both contained 2 MeNHX genes, while Chr01, Chr08, Chr09, Chr10 and Chr17 all contained only 1 MeNHX gene. The MeNHX protein had 520-1 157 amino acids, and the relative molecular mass and the theoretical isoelectric point were 58.09-128.21 kDa and 5.63-9.11, respectively. It belonged to amphoteric proteins with the unstable coefficient of 34.60-41.34 and hydrophobic protein with the GRAVY of 0.094-0.595. 9 MENHX genes contained typical Na~+/H~+ conserved domains. There were many synteny blocks in the M. esculenta NHX. The Ka/Ks values of evolutionary pressure all less than 1, and were mainly affected by purification selection. It was found that MeNHX gene, which contained the elements related to light response, biological stress and abiotic stress, had protein interaction with many ion channel genes.【Conclusion】In order to adapt to the change of environment, MeNHX gene joins the growth and development process of M. esculenta as it is influenced by light, biological and abiotic stress.
