使用响应面法优化酶动力学拆分(R/S)-δ-十四内酯
Optimizing the Enzymatic Kinetic Resolution of(R/S)-δ-Tetradecalactone by Response Surface Methodology
【摘要】 采用柱前衍生HPLC分析方法,以δ-十四内酯为底物,对一系列脂肪酶进行了筛选,得到可特异性拆分底物为(R/S)-5-羟基十四烷酸的黑曲霉脂肪酶(ANL)和洋葱假单胞菌脂肪酶(PCL).考察了pH、温度、助溶剂、酶酯比和反应时间对产物光学纯度(eep)、收率(Y)的影响.通过响应面分析方法 (CCD)确定了最佳反应条件,优化后得到(S)-5-羟基十四烷酸(eep1≥99.9%,收率为42.0%)和(R)-5-羟基十四烷酸(eep2≥99.9%,收率达49.0%).该研究符合绿色化学要求,具有良好的应用潜力.
【Abstract】 A series of lipases were screened using δ-tetradecalactone as substrate by pre-column derivatization HPLC analysis. Aspergillus niger lipase(ANL) and Pseudomonas cepacia lipase(PCL) with specific hydrolysis substrate of(R/S)-5-hydroxytetradecanoic acid were obtained. The effects of pH, temperature, cosolvent, enzymeester ratio and reaction time on the optical purity(eep) and yield(Y) of the product were investigated. The optimum reaction conditions were determined by response surface analysis(CCD). After optimization,(S)-5-hydroxytetradecanoic acid(eep1 ≥ 99.9%, yield 42.0%) and(R)-5-hydroxytetradecanoic acid(eep2 ≥ 99.9%, yield49.0%) were obtained. The system meets the requirements of green chemistry and has good application potential.
