猕猴桃根腐病菌的分离鉴定及不同种质的抗性分析

【摘要】 【目的】明确福建省三明市明溪县猕猴桃根腐病的病原菌种类,分析不同猕猴桃种质对该病原菌的抗性差异。【方法】采用组织分离法分离病原菌,通过形态学观察和分子生物学方法鉴定病原菌种类,根据科赫氏法则检测病原菌的致病性,并分析其对不同猕猴桃种质的致病差异。【结果】从猕猴桃典型根腐病组织分离纯化到1株病原菌株(编号0814-1),其菌落正面白色,背面呈白色或橘黄色,菌落边缘不规则。ITS和TEF序列的克隆测序及在NCBI数据库的比对结果显示,0814-1与轮纹镰孢菌(Fusarium concentricum)序列的同源性达99%以上。进化树分析结果亦表明,0814-1与轮纹镰孢菌聚在同一个进化分支。致病性测定结果表明0814-1引起的发病症状与田间症状一致,符合科赫氏法则。结合形态学、分子生物学和科赫氏法则测定结果,将分离到的0814-1病原菌鉴定为轮纹镰孢菌。猕猴桃种质对病原菌的抗性分析结果表明,不同种质对0814-1的抗性存在明显差异,其中‘金魁’和‘华特’为抗病种质。【结论】本研究分离鉴定到一种引起猕猴桃根腐病的病原菌轮纹镰孢菌,并筛选到2份抗病种质,为猕猴桃根腐病的综合防控及抗病育种奠定基础。

【Abstract】 【Objective】The type of pathogen causing the root rot of kiwifruits in Mingxi County,Sanming City,Fujian Province need to be clarified. And the differences in resistance to the pathogen among different kiwifruit germplasms also need to be studied.【Method】The pathogen was isolated by using tissue separation method. The morphological observation and molecular biological methods were used to identify the pathogen. Then,its pathogenicity was tested according to Koch’s postulates. And the resistance to the pathogen among different kiwifruit germplasms was analyzed.【Result】A pathogen(named 0814-1)was isolated from typical root rot tissues of kiwifruits. The colony was white on the surface,and white or orange on the back,with an irregular margin. The ITS an TEF were cloned from 0814-1 and sequenced,and the blast analysis in NCBI database showed that they shared more than 99% identity with that of Fusarium concentricum. The phylogenetic analysis also revealed that 0814-1 clustered with F. concentricum in the same branch. Pathogenicity test results showed that the symptoms caused by 0814-1 were consistent with that in the field,fulfilling Koch’s postulates. Based on the results of morphology,molecular biology,and Koch’s postulates,the isolated pathogen was identified as F. concentricum. Resistance analysis showed that there were significant differences in resistance to the pathogen 0814-1 among those germplasms,and‘Jinkui’and‘White’were screened as disease-resistant germplasms.【Conclusion】A pathogen causing kiwifruit root rot was isolated and identified as F. concentricum,then two disease-resistant germplasms were screened. Those results laid the foundation for the integrated control of kiwifruit root rot and resistance breeding.
