【摘要】 为了筛选高效裂解兔源致病性大肠埃希氏菌的宽谱噬菌体，用滴定法和双层平板法分离纯化出1株噬菌体vB-EcoS-tE10,用透射电镜观察该噬菌体的形态特征，并研究其生物学特性。结果表明，在透射电镜下其头部呈20面体，长径约为52.5 nm,横径约为51.6 nm,尾部长约86.9 nm,有明显的颈部。最佳感染复数为0.1,潜伏期为20 min,暴发期为80 min,裂解量为608 PFU/cell,在30～60℃作用0.5～1 h,其活性不受影响，在pH为6～9可保持很高的效价，能裂解9株兔源大肠埃希氏菌、1株鸡源大肠埃希氏菌和1株产肠毒素性大肠埃希氏菌K99,属于高效裂解性宽谱噬菌体，在大肠埃希氏菌噬菌体及裂解酶的开发和利用方面有较高价值。更多还原

【Abstract】 In order to screen a wide spectrum phage that can efficiently split pathogenic Escherichia coli from rabbits.The phage was isolated and purified by titration and double plate method,named vB-EcoStE10.The morphological characteristics of the phage were observed by transmission electron microscopy,then the biological characteristics were studied.The results showed that its head was an icosahedron with a long diameter of about 52.5 nm and a cross diameter of about 51.6 nm.Its tail length was about 86.9 nm with a distinct neck.The optimal complex number of infection was 0.1,the incubation period was 20 min,the eruption period was 80 min,and the burset size was about 608pfu/cell.In the temperature range of 30-60℃ for 0.5-1 h,the activity was almost not affected.In the range of pH 6-9,it can maintain a high titer.The phage can split 9 strains of rabbit Escherichia coli,1 strain of chicken Escherichia coli and 1 strain of enterotoxigenic Escherichia coli K99.It was an efficient lytic broad-spectrum phage,with great value in the development and utilization of Escherichia coli phage and its lyase.更多还原
