茄子再生体系的优化
Optimization of regeneration system of eggplant
【摘要】 为了建立茄子(Solanum melongena L.)高效再生体系,本试验选用11种不同类型茄子为试验材料,重点研究了基因型、外植体类型和激素种类与浓度配比对茄子再生效率的影响,优化了茄子的再生体系。结果表明:在11种不同基因型的茄子中,07-1043的再生能力最强,其次为16-562和14-345,其平均再生率分别为72.22%、48.33%、38.42%。我们对07-1043、16-562和14-345的子叶进行了形态和组织学分析发现,茄子再生能力的高低与子叶大小无明显相关性。ZRI(2 mg/L ZR+0.5 mg/L IAA)培养基对11种基因型茄子的再生苗诱导率最高。IAA浓度为0.1 mg/L时,对不定根的诱导效果最好。本研究优化了07-1043、16-562和14-345的离体再生体系,为茄子的基础理论研究和利用分子育种技术进行茄子种质创新助力。
【Abstract】 In order to establish an efficient regeneration system for eggplant(Solanum melongena L.), 11 genotypes of eggplant were used and the effects of genotypes, explant types as well as the types and concentration ratio of phytohormones on regeneration rate were studied. The results showed that among 11 different genotypes of eggplant,07-1043 had the highest regeneration ability, followed by 16-562 and 14-345, with average regeneration rates of 72.22%, 48.33% and 38.42%, respectively. In addition, morphology and histology analyses of cotyledons of the three genotypes were also performed, suggesting no obvious correlation between the size of cotyledon and the ability of regeneration. ZRI(2 mg/L ZR + 0.5 mg/L IAA) showed the best induction rate for seedlings regenation of the 11 genotypes. 0.1mg/L IAA showed the best effects on the induction of adventitious roots. This study optimized the regeneration systems of 07-1043, 16-562 and 14-345, which is helpful for the basic theoretical research of eggplant and the innovation of eggplant germplasm using molecular breeding technology.
