KASP-SNP标记鉴别白菜自交系结球或非结球性状的应用
Application of KASP-SNP markers to detecting heading or non-heading traits in Brassica inbred lines
【摘要】 为提高传统结球型或非结球型白菜的育种效率,提高遗传分析的准确性和育种的有效性。利用SNP分子标记,选取KBr43、KBr111、KBr233和KBr258共4套引物,对206份白菜自交系,包括148份结球型白菜和58份非结球型白菜分别进行了KASP分析。KASP分析结果表明,KBr111或KBr258 2套引物显示结球白菜与不结球白菜之间存在显著差异。在由3组引物KBr43、KBr111和KBr258标记的SNP位点组成的8个单倍基因型中,发现结球型白菜具有相对较高的ATG和ATC单倍基因型百分比,而非结球型白菜具有相对高的AGG或AGC单倍基因型比例。因此,我们推断,结球白菜更倾向于ATG和ATC单倍基因型,而非结球白菜更倾向于AGG和AGC单倍基因型。
【Abstract】 Leafy head was a significant agronomic trait of distinguishing heading type and non-heading type Chinese cabbage. The development of SNP molecular markers provided an auxiliary tool for leafy vegetable breeding, which could improve the efficiency of traditional heading or non-heading type Chinese cabbage, and greatly enhance the accuracy of genetic analysis and the effectiveness of breeding. In this study, 206 inbred lines of Brassica accessions,including 148 heading type and 58 non-heading type Chinese cabbages, were subjected respectively to KASP analysis using four sets of primers, including KBr43, KBr111, KBr233 and KBr258. There were significant differences of the KASP results between heading and non-heading Chinese cabbage using primers KBr111 or KBr258. Among the eight haplotypes composed of SNP sites labeled with three primers sets KBr43, KBr111 and KBr258, a relatively high percentage of ATG and ATC haplotypes was found in heading type Chinese cabbage, while non-heading type Chinese cabbage harbored a relatively high percentage of AGG or AGC haplotypes. Therefore, we inferred that heading Chinese cabbage was more inclined towards ATG and ATC haplotypes, with the difference being that non-heading Chinese cabbage was more tended to AGG and AGC haplotypes.
