棘孢小单孢菌DSM 43816双组分系统的生物信息学分析
The Bioinformatics Analysis of Two-Component Systems in Micromonospora echinospora DSM 43816
【摘要】 棘孢小单孢菌(Micromonospara echinospora)是一种能产生多种生物活性化合物的菌株,在药物发现和开发领域中具有重要的应用价值.双组分系统(two-component systems, TCSs)是存在于细菌内的一种信号传导系统,一般由组氨酸激酶(histidine kinase, HK)和应答调控蛋白(response regulator, RR)2部分组成,细菌可通过该系统感受外界环境变化,调控自身的生理过程以维持生存,是细菌适应选择压力的一种机制.该文以棘孢小单孢菌DSM 43816为研究对象,采用生物信息学方法系统分析了该菌株基因组内TCSs表达基因的分布、结构及功能.分析结果表明:棘孢小单孢菌DSM 43816基因组共编码35个组氨酸激酶和47个应答调控蛋白,其中包括21对配对的TCSs、2个杂合蛋白、14个孤儿HK蛋白、26个孤儿RR蛋白.这些配对的TCSs可能参与调控金属离子转运、硝酸盐和磷酸盐的代谢、生长运动、群体感应、应激反应、细菌毒力、细胞膜细胞壁的合成及抗生素的合成等多重生物学功能.研究结果可为揭示棘孢小单孢菌DSM 43816对环境的适应性机制、开发其生物活性化合物提供参考.
【Abstract】 Micromonospora echinospora is a strain of actinomycetes belonging to the genus Micromonospora which can produce various bioactive compounds and has important application value in drug discovery and development.Two-component systems(TCSs) refer to a signal transduction system that exists within bacteria, generally composed of two parts that are histidine kinase(HK) and response regulator(RR).Bacteria can sense changes in the external environment through this system, regulate their physiological processes to maintain survival, and are a mechanism by which they adapt to selection pressure.The study focuses on the distribution, structure, and function of TCSs contained in the genome of Micromonospora echinospora DSM 43816 using bioinformatics methods.The results show that there are a total of 35 HKs and 45 RRs in the genome of Micromonospora echinospora DSM 43816,including 21 paired TCSs, 2 hybrid HK-RR proteins, 14 orphan HKs and 26 orphan RRs. These paired TCSs may regulate multiple biological functions such as metal ion transport, nitrate and phosphate metabolism, growth movement, quorum sensing, stress response, bacterial virulence, synthesis of cell membrane and cell wall, and antibiotic synthesis.The above results can provide a reference for revealing the adaptive mechanism of Micromonospora echinospora DSM 43816 to the environment and developing its bioactive compounds.
