放射治疗对宫颈癌DNA甲基转移酶表达的影响
Effect of radiotherapy on the expression of DNA methyltransferase in cervical cancer
【摘要】 目的:研究放射治疗对宫颈癌(cervical cancer, CIN)组织中DNA甲基转移酶(DNA methyltransferase, DNMT)1、3A和3B表达的影响,探讨其在放射治疗耐受中的作用。方法:选用12例宫颈癌患者放射治疗前、中、后三个时期的组织,应用qPCR检测宫颈癌组织在放射治疗前期、中期和后期的DNMT1、DNMT3A和DNMT3B mRNA的表达变化。结果:宫颈癌放射治疗前、中、后三个时期DNMT1对β-actin mRNA的相对表达量分别是(0.122 9±0.021 7)、(0.569 6±0.021 7)和(0.162 0±0.035 2);放射治疗期间和放射治疗之前宫颈癌组织中DNMT1 mRNA的表达差异有统计学意义(P<0.05),放射治疗期间和放射治疗之后宫颈癌组织中DNMT1 mRNA的表达差异有统计学意义(P<0.05),放射治疗之前和放射治疗之后宫颈癌组织中DNMT1 mRNA的表达差异有统计学意义(P<0.05);DNMT3A对β-actin mRNA的相对表达量分别是(0.001 2±0.000 1)、(0.002 1±0.000 1)和(0.001 4±0.000 1),放射治疗期间和放射治疗之前宫颈癌组织中DNMT3A mRNA的表达差异有统计学意义(P<0.05),放射治疗期间和放射治疗之后宫颈癌组织中DNMT3A mRNA的表达差异有统计学意义(P<0.05),放射治疗之前和放射治疗之后宫颈癌组织中DNMT3A mRNA的表达差异无统计学意义(P>0.05);DNMT3B对β-actin mRNA的相对表达量分别是(0.662 7±0.034 8)、(0.810 4±0.084 5)和(0.239 1±0.047 0),放射治疗期间和放射治疗之前宫颈癌组织中DNMT3B mRNA的表达差异有统计学意义(P<0.05),放射治疗期间和放射治疗之后宫颈癌组织中DNMT3B mRNA的表达差异有统计学意义(P<0.05),放射治疗之前和放射治疗之后宫颈癌组织中DNMT3B mRNA的表达差异有统计学意义(P<0.05)。结论:放射治疗使宫颈癌组织中DNMT1,DNMT3A和DNMT3B表达发生变化,其可能在放射耐受中起到了作用。
【Abstract】 Objective:To investigate the effect of radiotherapy on the expression of DNA methyltransferase(DNMT) 1, 3A, and 3B in cervical cancer(CIN) tissues, and to explore its role in radiotherapy resistance. Methods:The cervical cancer tissues of 12 patients were selected before, during and after radiotherapy. The mRNA expression levels of DNMT1, DNMT3A and DNMT3B in cervical cancer tissues after radiotherapy were detected by qPCR. Results:The relative expression levels of DNMT1 to β-ACTIN were 0.122 9±0.021 7, 0.569 6±0.021 7 and 0.162 0±0.035 2 respectively before, during and after radiotherapy for cervical cancer. The expression of DNMT1 mRNA in cervical cancer tissues during and before radiotherapy was significantly different(P<0.05), and the expression of DNMT1 mRNA in cervical cancer tissues during and after radiotherapy was significantly different(P<0.05). The expression of DNMT1 mRNA in cervical cancer tissues before and after radiotherapy was significantly different(P<0.05). The relative expression levels of DNMT3A to β-ACTIN mRNA were 0.001 2±0.000 1, 0.002 1±0.000 1 and 0.001 4±0.000 1 respectively. There were significant differences in DNMT3A mRNA expression in cervical cancer tissues during radiotherapy and before radiotherapy(P<0.05). The expression of DNMT3A mRNA in cervical cancer tissues during and after radiotherapy was significantly different(P<0.05), however, the expression of DNMT3A mRNA before and after radiotherapy was not significantly different(P>0.05). The relative expression of DNMT3B on β-ACTIN mRNA was 0.662 7±0.034 8, 0.810 4±0.084 5 and 0.239 1±0.047 0 respectively. The mRNA expression of DNMT3B was significantly different in cervical cancer tissues during and before radiotherapy(P<0.05). The expression of DNMT3B mRNA in cervical cancer tissues during and after radiotherapy was significantly different(P<0.05), and the expression of DNMT3B mRNA in cervical cancer tissues before and after radiotherapy was significantly different(P<0.05). Conclusion:Radiotherapy changes the expression of DNMT1, DNMT3A and DNMT3B in cervical cancer tissues, which may play an important role in radioresistance.
