硝酸钾预处理对刺五加种子形态后熟的调节机制
Regulatory Mechanism of Potassium Nitrate Pretreatment on Morphological Post-ripening of Seeds of Acanthopanax senticosus
【摘要】 以刺五加(Acanthopanax senticosus(Rupr.Maxim.) Harms)野生种子为试材,经不同浓度的硝酸钾(KNO3)溶液浸种处理48 h后,在18~22℃条件下进行层积处理,测定和分析了种子形态后熟期间的种子胚率、腐烂率和相关生理生化指标,以期为刺五加简化育苗提供参考依据。结果表明:KNO3预处理显著提高了硝酸还原酶(NR)和淀粉酶活性,增加了一氧化氮(NO)和可溶性糖含量,促进了刺五加种胚的形态后熟;同时,调控了抗氧化酶活性水平,缓解了活性氧(ROS)对细胞的损伤,降低了丙二醛(MDA)含量,减少了腐烂率。因此,KNO3预处理可有效促进刺五加种子形态后熟,缩短高温层积时间,降低层积期间种子的腐烂率。
【Abstract】 The wild seeds of A.senticosus were pre-soaked with different concentrations of potassium nitrate(KNO3) solution for 48 hours, and then stratified at 18-22 ℃.The embryo rate, decay rate, and related physiological and biochemical indexes of the seeds were determined during the stacking process, in order to provide reference for the simplified cultivation of A.senticosus.The results showed that KNO3 pre-treatment significantly improved the activities of nitrate reductase and amylase, increased nitric oxide and soluble sugar content, and promoted the morphological post-ripening of seed embryo of A.senticosus. At the same time, the level of antioxidant enzyme activity was regulated, and the damage of active oxygen(ROS) to cells was alleviated and the content of MDA and decay rate of seeds was reduced.In conclusion, KNO3 pretreatment could effectively promote the morphological maturation of A.senticosus seeds, shorten the time of high-temperature stratification, and reduce the rate of seed deterioration during stratification.
