补中益气汤加减升麻、柴胡对脾虚泄泻大鼠胃肠、免疫功能及肠黏膜屏障的影响差异
Effects of Buzhong Yiqi Decoction Modified with Cimicifugae Rhizoma,Bupleuri Radix on Gastrointestinal Function,Immune Function and Intestinal Mucosal Barrier in Splenasthenic Diarrhea Rats
【摘要】 目的 研究引经药升麻、柴胡对补中益气汤治疗脾虚泄泻的引经增效作用。方法 60只雄性SD大鼠随机分为正常组、模型组、补中益气汤全方组、补中益气汤缺升麻组、补中益气汤缺柴胡组和补中益气汤缺升麻柴胡组。除正常组外,其余大鼠采用利血平~+大黄~+隔日进食法复制脾虚泄泻大鼠模型。从第3周开始,各组分别灌胃相应药物,正常组和模型组灌胃等体积蒸馏水,连续给药2周。末次给药后,计算腹泻、脾脏及胸腺指数;苏木素-伊红(HE)法观察空肠组织病理学变化;酶联免疫吸附法(ELISA)检测血清中白细胞介素6(IL-6)、白细胞介素10(IL-10)、胃泌素(GAS)、血管活性肠肽(VIP)水平;流式细胞术检测外周血T淋巴细胞亚群CD3~+、CD4~+、CD8~+T淋巴细胞百分比;Western Blot法检测空肠中p38丝裂原活化蛋白激酶(p38MAPK)、磷酸化p38丝裂原活化蛋白激酶(p-p38 MAPK)、闭锁小带蛋白1(ZO-1)、闭合蛋白(Occludin)的表达。结果 与正常组比较,模型组大鼠腹泻指数、IL-6水平及p-p38 MAPK蛋白表达明显升高(P<0.01),脾脏及胸腺指数、血清IL-10、GAS、VIP、CD3~+、CD4~+、CD4~+/CD8~+比值、空肠ZO-1、Occludin蛋白表达均明显降低(P<0.01),空肠可见黏膜损伤。与模型组比较,补中益气汤全方组给药后大鼠腹泻指数、IL-6水平、p-p38 MAPK蛋白表达明显降低(P<0.05,P<0.01),脾脏及胸腺指数、血清IL-10、GAS、VIP、CD3~+、CD4~+、CD4~+/CD8~+比值、空肠ZO-1、Occludin蛋白表达明显增高(P<0.05,P<0.01),空肠黏膜损伤不同程度地改善,较其他组改善更明显。结论 升麻、柴胡具有增强补中益气汤改善脾虚泄泻大鼠胃肠和免疫功能,修复肠黏膜损伤的效应。
【Abstract】 Objective To study the synergized action through meridian guiding of Cimicifugae Rhizoma,Bupleuri Radix combined with Buzhong Yiqi Decoction(BZYQ) on the treatment of splenasthenic diarrhea.Methods Sixty male SD rats were randomly divided into normal group, model group, BZYQ group, BZYQ group with out Cimicifugae Rhizoma deficiency, BZYQ group with out Bupleuri Radix, and BZYQ group with out Cimicifugae Rhizoma and Bupleuri Radix. Except the normal group,rats in other groups were treated with Reserpine~+Rhei Radix et Rhizoma~+alternate-day feeding to establish a rat model with splenasthenic diarrhea. Each treatment group was given the corresponding drugs respectively,and the normal group and the model group were given the same volume of distilled water(starting from the third to the fifth week). After the last administration,diarrhea index,spleen index and thymus index were calculated. Hematoxylin-eosin(HE) method was used to observe the pathological changes of jejunum tissue. Enzyme-linked immunosorbent assay(ELISA) method was used to analyze the contents of interleukin-6(IL-6),interleukin-10(IL-10),gastrin(GAS) and vasoactive intestinal peptide(VIP) in serum. Flow cytometry was used to detect the percentages for T-lymphocyte subpopulations,including CD3~+,CD4~+,CD8~+in peripheral blood. Western Blot was used to detect the expressions of p38 mitogen-activated protein kinase(p38 MAPK),phosphorylated p38 mitogen-activated protein kinase(p-p38 MAPK),zonula occludens-1(ZO-1)and occludin protein(Occludin) in jejunum.Results Compared with the normal group,the diarrhea index,the level of IL-6 and p-p38 MAPK in the model group were significantly increased(P<0.01),spleen index,thymus index, serum IL-10, GAS, VIP, CD3~+and CD4~+cells, CD4~+/CD8~+ratio, protein expressions of ZO-1 and Occludin in jejunum were significantly decreased(P<0.01),the epithelial mucosal of the jejunum was damaged.Compared with the model group, the diarrhea index, the level of IL-6 and p-p38 MAPK were significantly decreased(P<0.05,P<0.01) after administration of BZYQ. Spleen index,thymus index,serum IL-10,GAS,VIP, percentages of CD3~+and CD4~+cells, CD4~+/CD8~+ratio, and protein expressions of ZO-1 and Occludin in jejunum were significantly increased(P<0.05,P<0.01). A significant improvement of jejunal mucosal injury was observed in BZYQ group.Conclusion Cimicifugae Rhizoma and Bupleuri Radix have the enhancing effects for improving gastrointestinal, immune function and repairing intestinal mucosal damage of BZYQ in splenasthenic diarrhea rats.
