基于网络药理学探讨左金丸治疗功能性消化不良作用机制
Exploration on the Mechanism of Zuojin Pills in Treating Functional Dyspepsia Based on Network Pharmacology
【摘要】 目的 基于网络药理学分析左金丸治疗功能性消化不良(FD)的作用靶点和分子机制。方法 检索中药系统药理学数据库与分析平台(TCMSP)获得左金丸的有效成分及作用靶点,检索GeneCards数据库获取FD的疾病靶点,并使用韦恩图获取药物与疾病的共同靶点。采用STRING数据库进行蛋白质-蛋白质相互作用(PPI)分析,使用Cytoscape 3.7.1进行PPI网络拓扑分析。使用Metascape数据库进行GO功能和KEGG通路富集分析。采用Cytoscape 3.7.1构建“药物-有效成分-关键靶点-通路”网络。结果 获得左金丸33个有效成分,作用靶点203个,其关键有效成分为槲皮素、异鼠李素、β-谷甾醇、吴茱萸次碱、氢化小檗碱等,关键靶点为AKT1、TP53、TNF、IL6、VEGFA。GO功能富集到1 989个条目,KEGG通路富集到202条通路,主要包括癌症通路、脂质与动脉粥样硬化、PI3K-Akt信号通路、乙型肝炎等。结论 左金丸治疗FD具有多成分、多靶点、多通路的特点,通过调控AKT1、TP53、TNF、IL6、VEGFA等核心靶点和PI3K-Akt信号通路等,发挥抗凋亡、抗炎、抗抑郁、改善胃肠功能等作用。
【Abstract】 Objective To analyze the targets and mechanism of Zuojin Pills in treating functional dyspepsia(FD) based on network pharmacology. Methods The effective components and targets of Zuojin Pills were obtained by retrieving TCMSP, and the disease targets of FD were obtained by searching GeneCards database. The common targets of drugs and diseases were obtained by using Wayne diagram. Protein-protein interaction(PPI) analysis was conducted using STRING database. PPI network topology analysis was conducted using Cytoscape 3.7.1. GO function and KEGG pathway enrichment analysis were performed using Metascape database. Cytoscape 3.7.1 was used to construct the “drug-effective components-key targets-pathway” network. Results Totally 33 effective components and 203 targets of Zuojin Pills were obtained. The key effective components were quercetin,isorhamnetin, β-sitosterol, rutaecarpine and hydrogenated tiller base. The key targets were AKT1, TP53, TNF, IL6and VEGFA. GO enriched 1 989 entries, and KEGG enriched 202 pathways, including cancer pathways, lipid and atherosclerosis, PI3K-Akt signaling pathway, hepatitis B, etc. Conclusion Zuojin Pills has the characteristics of multicomponent, multi-target and multi-pathway in the treatment of FD. It regulates AKT1, TP53, TNF, IL6, VEGFA and other core targets and PI3K-Akt signaling pathway and other pathways to play the roles of anti-apoptosis, antiinflammation, anti-depression and improving gastrointestinal function.
