化浊行血汤对高脂血症大鼠脂代谢及肝组织二脂酰甘油酰基转移酶2 mRNA表达的影响

【摘要】 目的:分析化浊行血汤对高脂血症模型大鼠脂代谢及肝组织二脂酰甘油酰基转移酶2(Diacylglycerol Acyltransferase 2,DGAT2)mRNA表达的影响。方法:选取SD大鼠70只,分为空白组、模型组、脂必妥组、辛伐他汀组、中药组,其中,中药组又具体划分为高、中、低3个剂量组,除空白组外均给予高脂饮食制备高脂血症大鼠模型。造模成功后给予药物治疗8周,检测血清总胆固醇(Total Cholesterol,TC)、载脂蛋白A1(Apolipoprotein A1,Apo-A1)等;实时荧光定量聚合酶链式反应法检测大鼠肝组织DGAT2 mRNA的表达水平。结果:模型组大鼠血清TC、三酰甘油(Triglycerides,TG)、低密度脂蛋白胆固醇(Low-density Lipoprotein Cholesterol,LDL-C)水平均高于空白组,高密度脂蛋白胆固醇(High-density Lipoprotein Cholesterol,HDL-C)、Apo-A1水平低于空白组,且两组数据差异较大(P <0.01);此外,经过检测发现,相较于模型组,各治疗组血清TC、TG、LDL-C含量均减少,与此同时,其他组的HDL-C、Apo-A1水平均提升,且对比可知以上指标差异较大(P <0.05,P <0.01);在中药组中,采用药物干预一段时间后,中剂量组较另外两组TC、TG下降,而HDL-C相应增加,组间对比结果显示差异显著(P <0.05)。在DGAT2 mRNA表达方面,模型组较空白组含量显著增加,而与其他组相比又呈现出下降趋势(P <0.05);在中药组中,中剂量组较另外两组均明显减少,且差异显著(P <0.05)。结论:化浊行血汤可起到调节脂质代谢的效用,本次研究得出,其功效或与下调肝脏DGAT2 mRNA的表达有关。

【Abstract】 Objective: To analyze the effects of the Huazhuo Xingxue decoction(化浊行血汤) on lipid metabolism and diacylglycerol acyltransferase 2(DGAT2) mRNA expression in liver of hyperlipidemia model rats. Methods: A total of 70 SD rats were selected and divided into the blank group, the model group, the Zhibituo(脂必妥) group, the simvastatin group, and the TCM medicine group; the TCM medicine group was specific ally divided into high, medium and low dose groups. Except for the blank control group, others groups were given a high-fat diet to establish hyperlipidemia rat model. After the success of preparation, medcine treatment was given for 8 weeks, serum total cholesterol(TC) and apolipoprotein A1(Apo-A1) were detected. RT-PCR was used to detect the mRNA expression of DGAT2 in rat liver tissue. Results: The serum TC, triglycerides(TG) and low-density lipoprotein cholesterol(LDL-C) levels in the model group were higher than those in the blank group, while the levels of high-density lipoprotein cholesterol(HDL-C) and Apo-A1 were lower than those in the blank group, and the data in the two groups was significantly different(P<0.01). In addition, compared with the model group, serum TC, TG and LDL-C contents in all treatment groups were decreased, while HDL-C and Apo-A1 levels in other groups were increased, and the above indicators were significantly different(P<0.05, P<0.01). In the TCM medicine group, TC and TG were decreased in the medium-dose group compared with the other two groups after drug intervention for a period of time. HDL-C was increased, and the difference between groups was significant(P<0.05). DGAT2 mRNA expression in the model group was significantly higher than that in the blank group. Compared with other groups, it showed a downward trend(P<0.05). In the TCM medicine group, the DGAT2 mRNA level in the medium-dose group was significantly lower than the other two groups, and the difference was significant(P<0.05). Conclusion: The Huazhuo Xingxue decoction can regulate lipid metabolism, which may be related to the downregulation of liver DGAT2 mRNA expression in this study.
