全三七片对血管内皮细胞损伤的保护作用研究

【摘要】 目的 观察全三七片(PSQ)对炎症和脂质过氧化引起的血管内皮损伤的保护作用。方法 以人脐静脉内皮细胞(ECV304)为研究对象,分别采用炎症介质肿瘤坏死因子(tumor necrosis factor, TNF)-α和氧化低密度脂蛋白(oxidized low density lipoprotein, ox-LDL)诱导ECV304血管内皮细胞建立损伤模型,设正常对照组、模型对照组、阳性药组和PSQ不同浓度组,检测血管内皮功能指标一氧化氮(nitric oxide, NO)、总一氧化氮合成酶(total nitric oxide synthase, T-NOS)和内皮素(endothelin,ET)-1,氧化应激相关指标丙二醛(malondialdehyde, MDA)和超氧化物歧化酶(superoxide dismutase, SOD)和炎症相关指标白介素(interleukin, IL)-1β、IL-6和TNF-α。结果 TNF-α造模24 h后,与正常对照组比较,模型对照组细胞上清中的ET-1、NO、MDA、IL-1β和IL-6水平显著升高(P<0.05或P<0.01),SOD水平显著降低(P<0.05);与模型对照组比较,PSQ不同浓度组细胞上清中的ET-1、NO、MDA、IL-1β和IL-6水平显著降低(P<0.05或P<0.01),SOD水平显著升高(P<0.01)。ox-LDL造模24 h后,与正常对照组比较,模型对照组细胞上清中的ET-1、MDA、IL-1β和TNF-α水平显著升高(P<0.01);与模型对照组比较,PSQ不同浓度组细胞上清中的NO、MDA、T-NOS、ET-1、IL-1β和TNF-α水平显著降低(P<0.05或P<0.01),SOD水平显著升高(P<0.05或P<0.01)。结论 全三七片可能通过改善氧化应激反应、保护血管内皮功能以及减少炎症因子的分泌进而保护炎症和脂质过氧化诱导的血管内皮损伤。

【Abstract】 Objective To observe the protective effect of Quan Sanqi tablets(PSQ) on vascular endothelial injury caused by inflammation and lipid peroxidation. Methods Human umbilical vein endothelial cells(ECV304) were used as the study object, and the injury model of ECV304 vascular endothelial cells was established by the inflammatory mediators’ tumor necrosis factor(TNF)-α and oxidized low density lipoprotein(ox-LDL), respectively. Normal control group, model control group, positive drug group, and PSQ groups with different concentrations were set up. Nitric oxide(NO), total nitric oxide synthase(T-NOS) and endothelin(ET)-1, malondialdehyde(MDA) and superoxide dismutase(SOD), and interleukin(IL)-1β, IL-6 and TNF-α were detected. Results After 24 hours of TNF-α modeling, compared with the normal group, the levels of ET-1, NO, MDA, IL-1β and IL-6 in the supernatant of the model group were significantly increased(P<0. 05 or P<0. 01), and the level of SOD was significantly decreased( P< 0. 05). Compared with the model group, PSQ could significantly decrease the levels of ET-1, NO, MDA, IL-1β, and IL-6 in the supernatant(P< 0. 05 or P< 0. 01), and increased the level of SOD(P<0. 01). After 24 h of ox-LDL modeling, the levels of ET-1, MDA, IL-1β, and TNF-α in the supernatant of the model group were significantly increased compared with the normal group(P<0. 01). Compared with the model group, PSQ significantly decreased the levels of NO, TNOS, ET-1, MDA, IL-1β, and TNF-α in the supernatant(P<0. 05 or P<0. 01), and increased the level of SOD(P<0. 05 or P<0. 01). Conclusion Quan Sanqi tablets may protect against vascular endothelial injury by improving oxidative stress response, protecting vascular endothelial function, and reducing the secretion of inflammatory factors.
