miR-25通过调节TGF-β信号传导通路促进胃癌细胞增殖和转移
miR-25 Promotes Proliferation and Transfer of Gastric Cancer Cell by Regulating TGF-β Signaling Pathway
【摘要】 目的:探究miR-25对人胃癌细胞SGC-7901增殖、侵袭、迁移、凋亡的作用及其分子机制。方法:构建miR-25干扰慢病毒,并将研究目的细胞分为正常组、miR-25干扰慢病毒组和miR-25干扰慢病毒空载组,利用现代医学生物学技术,检测转染效果、细胞增殖能力、侵袭能力、迁移能力、细胞凋亡情况,以及各组细胞中转化生长因子-β(TGF-β)蛋白的表达情况。结果:与正常组相比,miR-25在mi R-25干扰慢病毒组中的表达明显更低(P<0.05);与正常组相比,miR-25干扰慢病毒组的细胞存活率、迁移率、增殖率、侵袭能力均显著降低,凋亡率明显更高,差异均有统计学意义(P<0.05)。与正常组相比,TGF-β蛋白在miR-25干扰慢病毒组中的表达量增加,差异有统计学意义(P<0.05)。结论:miR-25在人胃癌细胞SGC-7901中的表达被抑制后,TGF-β蛋白在胃癌细胞中的表达显著增加,且明显降低了胃癌细胞的侵袭转移能力,并促进了肿瘤细胞的凋亡。
【Abstract】 Objective: To explore the effects of miR-25 on proliferation, invasion, migration and apoptosis of human gastric cancer cell SGC-7901 and its molecular mechanism. Method: miR-25 interfering lentivirus was constructed, and the cells for study were divided into normal group, miR-25 interfering lentivirus group and miR-25interfering lentivirus no-load group. Modern medical biology technology was used to detect transfection efficiency,cell proliferation ability, invasion ability, migration ability, apoptosis situation, and the expression of transforming growth factor-β(TGF-β) protein in each group. Result: Compared with the normal group, the expression of miR-25in miR-25 interfering lentivirus group was significantly lower(P<0.05). Compared with the normal group, the survival rate, migration rate, proliferation rate and invasion ability of cell in miR-25 interfering lentivirus group were significantly decreased, and the apoptosis rate in miR-25 interfering lentivirus group was significantly higher, the differences were statistically significant(P<0.05). Compared with the normal group, the expression level of TGF-βprotein in miR-25 interfering lentivirus group was increased, the difference was statistically significant(P<0.05).Conclusion: After the expression of miR-25 was inhibited in human gastric cancer cell SGC-7901, the expression of TGF-β protein in gastric cancer cell was significantly increased, and the invasion and metastasis ability of gastric cancer cell were significantly reduced, and the apoptosis of tumor cells was promoted.
