泡核桃离体种胚快繁体系建立
Establishment of in vitro seed embryo rapid propagation system of Juglans sigillata
【摘要】 以泡核桃(Juglans sigillata)‘黔核7号’种胚为外植体,成功建立离体种胚快繁体系。结果表明,缓慢生长期的种胚萌发效果优于成熟期,以DKW+1.0 mg·L-1 6-苄基腺嘌呤(6-BA)+0.1 mg·L-1吲哚-3-丁酸(IBA)培养基最为适宜;适合种胚单芽增殖的培养基配方为DKW+2.0 mg·L-1 6-BA+0.1 mg·L-1 IBA+0.02mg·L-1吲哚-3-乙酸(IAA),增殖系数达11.23;适宜试管苗单芽茎段增殖的培养基配方为DKW+0.99 mg·L-16-BA+0.01 mg·L-1 IBA,增殖系数达8.03。采用两步生根法,即泡核桃试管苗在附加5.0 mg·L-1 IBA的1/4DKW培养基中黑暗培养12 d,随后转移到1/4DKW+120 g·L-1珍珠岩+20 g·L-1蔗糖培养基中光照生根,生根率为72.21%;生根试管苗采用珍珠岩和营养土两步炼苗,成活率达到92.48%。
【Abstract】 The rapid propagation system of in vitro seed embryos of Juglans sigillata ‘Qianhe 7’ as explant was successfully established in our study. The results show that the germination rate of seed embryo in slow growth stage was higher than in mature stage, and the optimal medium was DKW + 1.0 mg·L-1 6-benzylaminopurine(6-BA) + 0.1 mg·L-1 indole 3-butyric acid(IBA). The DKW + 2.0 mg·L-1 6-BA + 0.1 mg·L-1 IBA +0.02 mg·L-1 3-indoleacetic acid(IAA) medium could produce the highest bud proliferation rate of J. sigillata embryo which could reach 11.23. The suitable medium for the proliferation of single bud stem segments of test-tube plantlets was DKW + 0.99 mg·L-1 6-BA + 0.01 mg·L-1 IBA, and the proliferation coefficient was up to 8.03. In order to develop the root system, two-step rooting method was adopted. The testtube plantlets were cultured in dark in 1/4DKW medium with 5.0 mg·L-1 IBA for 12 days, then transferred to 1/4DKW + 120 g·L-1 perlite + 20 g·L-1 sucrose; the rooting rate was 72.21%. The rooting test-tube plantlets were refined with perlite and nutritious soil, and the survival rate reached 92.48%.
