麝香心脑乐片薄膜衣片减轻大鼠冠状动脉粥样硬化斑块负荷的机制研究
Mechanism of Shexiang Xinnaole Film-Coated Tablets alleviating atherosclerotic plaque burden in rats
【摘要】 目的 探讨麝香心脑乐片薄膜衣片对冠状动脉粥样硬化斑块负荷的影响及作用机制。方法 90只大鼠随机分为3组,每组30只,包括模型组(高脂饲料+0.9%氯化钠盐溶液)、实验组(高脂饲料+麝香心脑乐薄膜衣片溶液)、空白对照组(普通词料饲养+0.9%氯化钠盐溶液),全部大鼠造模成功后连续灌胃8周。HE染色检测冠脉斑块大小及泡沫细胞含量,生化检测血清中总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)、高密度脂蛋白胆固醇(HDL-C)水平,ELISA法检测血清中白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)水平。结果 麝香心脑乐片薄膜衣片降低了大鼠动脉粥样硬化斑块负荷及泡沫细胞数量,降低了冠脉粥样硬化大鼠血清中TC、TG、IL-1β、TNF-α水平(P<0.05)。结论 麝香心脑乐薄膜衣片可能通过减轻大鼠的脂质及炎症水平减轻冠状动脉粥样硬化斑块负荷。
【Abstract】 Objective To investigate the effect of Shexiang Xinnaole Film-Coated Tablets on atherosclerotic plaque burden in rats and its mechanisms.Methods 90 rats were randomly divided into 3 groups with 30 rats in each group, including model group(high-fat diet+0.9% sodium chloride salt solution),experimental group(high-fat diet+Shexiang Xinnaole Film-Coated Tablets solution),control group(normal food+0.9% sodium chloride salt solution).All rats were continuously gavaged for 8 weeks.The size of coronary plaque and the number of foam cells are detected by HE staining.The levels of serum total cholesterol(TC),triglyceride(TG),LDL-C and HDL-C are detected by biochemical analysis.The levels of serum interleukin-6(IL-6) and tumor necrosis factor-α(TNF-α) were detected by enzyme linked immunosorbent assay(ELISA).Results The new formulation of Shexiang Xinnaole Film-Coated Tablets reduced the atherosclerotic plaque burden, the number of foam cells and the levels of serum TC、TG、IL-1β、TNF-α in atherosclerotic rats(P<0.05).Conclusion The new formulation of Shexiang Xinnaole Film-Coated Tablets may reduce coronary atherosclerotic plaque burden by reducing lipid and inflammatory levels in rats.
