携带遗传标记的新型鹅细小病毒感染樱桃谷北京鸭再现短喙与矮小综合征
Reproduction of short beak and dwarfism syndrome in Cherry Valley Pekin ducks infected with rescued novel goose parvovirus carrying a genetic marker
【摘要】 为了阐明是否新型鹅细小病毒(novel goose parvovirus, NGPV)单独感染樱桃谷北京鸭能够复制出短喙与矮小综合征(short beak and dwarfism syndrome, SBDS)的所有临床症状,并进一步解析NGPV致病机理。本研究中,基于NGPV分离株SDJN19,构建了携带遗传标记的感染性克隆质粒pJNm。pJNm质粒转染9日龄樱桃谷北京鸭胚拯救出感染性病毒rJNm。rJNm半数致死量(ELD50)达到5×106.25/mL,与亲本株接近。rJNm感染2日龄樱桃谷北京鸭,能够复制出SBDS的所有典型临床特征,包括短喙、舌头外伸、生长迟缓、站立行走困难等。试验还表明rJNm能够通过水平传播途径感染樱桃谷北京鸭,感染鸭依旧表现出典型的SBDS特征。水平接触组鸭在感染后31 d能够同时在体内检测出高的病毒载量和高水平血清沉淀抗体,预示着NGPV可以持续性感染樱桃谷北京鸭。本研究结果表明SBDS确系NGPV感染所致,为研制针对性的预防性生物制剂以及深入解析NGPV的致病机制奠定了坚实基础。
【Abstract】 To elucidate whether sole novel goose parvovirus(NGPV) infection of Cherry Valley Pekin ducks can reproduce all the typical clinical symptoms of short beak and dwarfism syndrome(SBDS) and unveil the NGPV pathogenesis, a plasmid pJNm was constructed, which contained the entire genome of the NGPV isolate SDJN19 and carried a genetic marker.Transfection of pJNm in 9-day-old embryonated Cherry Valley Pekin duck eggs resulted in rescue of the infectious virus, named rJNm.rJNm had a medium embryo lethal dose(ELD50)of 5×106.25/mL,which was similar to the ELD50 value of the parental strain SDJN19(5×105.38/mL).rJNm infection of 2-day-old Cherry Valley Pekin ducks reproduced all characteristic signs of SBDS,including beak atrophy, tongue protrusion, retarded growth and difficulty in standing and moving.rJNm could invade ducks via the horizontal transmission route and the infected ducks still manifested the characteristic signs of SBDS.High levels of viral load and high titers of serum precipitation antibody could be simultaneously detected in the horizontal contact group ducks,demonstrating the persistent NGPV infection in ducks.Taken together,the present findings demonstrate that NGPV is the truly causative agent of SBDS,and lay a solid foundation for preparing the prophylactic biological agent and exploring the NGPV pathogenesis.
