丹参多糖对氟苯尼考处理的雏鸡肝脏中PPAR信号通路的调节作用

【摘要】 将120只雏鸡随机平均分为空白对照组、氟苯尼考(florfenicol, FFC)组和FFC+丹参多糖(Salvia miltiorrhiza polysaccharides, SMPs)组。从1日龄起,连续给药5 d。于第6天采取肝脏样品,进行转录组和蛋白组测序;采集和制备血清样品,保存剩余肝组织,用于检测其他相关的试验指标。采用qPCR和PRM的方法验证测序结果。测序和验证结果显示,暴露于FFC之后,雏鸡肝脏中1 989个基因和917个蛋白质发生了显著变化。这些显著改变了的基因和蛋白与对刺激的反应、异生物质的代谢与氧化解毒等生物功能有关。FFC显著改变了PPAR信号通路中的13个基因和18个蛋白,SMPs的干预则调整了其中的7个显著差异基因和8个显著差异蛋白的表达水平。此外,FFC还显著地提高了雏鸡血清中AST和ALT的含量,降低了肝脏中TG、TC和ANDP的含量。SMPs则有效逆转了以上因子的变化趋势。结果表明,FFC改变了PPAR信号通路关键因子的表达水平,进而造成了雏鸡肝脏的脂质代谢紊乱。SMPs则通过进一步调控该通路,在一定程度上改善了雏鸡肝脏的脂质代谢紊乱,进而维护了肝脏的正常功能。

【Abstract】 In order to study the directional regulation of Salvia miltiorrhiza polysaccharides(SMPs) on the specific signal pathway changed by florfenicol(FFC) in chick liver and the interaction between specific key genes and proteins in the pathway, 120 chicks were randomly and averagely divided into the control group, FFC group and FFC+SMPs group.From one day of age, the drug was administered continuously for five days.On the 6th day, liver samples were taken for transcriptome and proteome sequencing.The serum samples were collected and prepared, and the remaining liver tissue was preserved for the detection of other relevant indexes.qPCR and PRM were used to verify the sequencing results.Sequencing and validation results showed significant changes in 1 989 genes and 917 proteins in the liver of chicks exposed to FFC.These significantly altered genes and proteins were related to biological functions such as response to stimulation, metabolism of xenobiotics and oxidative detoxification.FFC significantly changed 13 genes and 18 proteins in PPAR signaling pathway,and the intervention of SMPs adjusted the expression levels of 7 significantly different genes and 8 significantly different proteins.In addition,FFC significantly increased the contents of AST and ALT in serum and decreased the contents of TG,TC and ANDP in liver.SMPs effectively reversed the change trend of the above factors.It is concluded that FFC changed the expression levels of key factors in PPAR signaling pathway,which leaded to the disorder of lipid metabolism in chick liver.By further regulating this pathway,SMPs improved the disorder of lipid metabolism in chick liver to a certain extent,and then maintained the normal function of liver.
