纳米氧化锌抑制柔嫩艾美耳球虫感染的作用研究

【摘要】 为探究纳米氧化锌(ZNP)对鸡球虫病的疗效。将180只13日龄的雏鸡随机分为对照组(Con),模型组(Mod),ZNP低剂量组(ZNP-10,10 mg/kg)、ZNP高剂量组(ZNP-20,20 mg/kg),地克珠利组(Dic, 1 mg/kg),ZNP药物对照组(Dcp, 20 mg/kg)。试验以6×10~4个/mL柔嫩艾美耳球虫(E.tenella)孢子化卵囊经口感染15日龄雏鸡(1 mL/只)建立鸡柔嫩艾美耳球虫感染动物模型,并在13日龄开始灌胃给药,每日1次,连续8 d。试验结束时,采集盲肠组织,进行病变评分;采用RT-qPCR及ELISA的方法检测盲肠扁桃体中炎性因子的表达量。结果显示,Mod组出现大量血便,剖检可见盲肠芯,盲肠组织切片中可见虫体,出血明显;ZNP-20组改善了球虫感染的特征病变,抗球虫指数(Anticoccidial index, ACI)达到159;相较于Mod组,ZNP能够降低IL-1β和TNF-α mRNA水平,提高GM-CSF、TGF-β、sIgA、IgM和IgG的蛋白表达(P<0.01)。结果表明,20 mg/kg ZNP具有一定的抗球虫作用,可能与其调节肠道黏膜免疫和炎症反应有关。

【Abstract】 The aim of this study is to investigate the effect of nano zinc oxide(ZNP) on chicken coccidiosis.One hundred and eighty 13-day-old chickens were randomly divided into the control group(Con),model group(Mod),ZNP low-dose group(ZNP-10,10 mg/kg),ZNP high-dose group(ZNP-20,20 mg/kg),Diclazuril group(Dic, 1 mg/kg) and ZNP drug control group(Dcp, 20 mg/kg).In the experiment, 15-day-old chicks(1 mL/chick) were orally infected with sporulated oocysts of E.tenella at 6×10~4/mL to establish an animal model of E.tenella infection in chicken, and gavage administration was started at 13-day-old once a day for seven consecutive days.At the end of the experiment, cecum tissue was collected, pathological changes were observed and scored.RT-qPCR and ELISA were used to detect the expression of inflammatory factors in amygdala caecum.The results showed that the sick chickens infected with coccidiosis had many blood stools with cecal cores, and the histopathological observation showed a lot of oocysts and obvious bleeding.ZNP-20 group improved the characteristic lesions of coccidial infection and the anticoccidial index(ACI) was 159.Compared with Mod group, ZNP could reduce the mRNA expression levels of IL-1β and TNF-α and up-regulated the proteins expressions of GM-CSF,TGF-β,sIgA,IgM and IgG(P<0.01).20mg/kg ZNP has a certain anticoccidial effect,which may be related to the regulation of intestinal mucosal immunity and inflammatory response.
