基于UPLC-MS/MS羊肚菌子实体酚酸类代谢物差异分析
Differential analysis of phenolic acid metabolites in fruitbody of Morchella esculenta based on UPLC-MS/MS
【摘要】 为了探究羊肚菌子实体酚酸类代谢物差异,采用超高液相色谱-串联质谱(UPLC-MS/MS)技术分析羊肚菌MLM和MTL子实体中酚酸类化合物种类及含量,利用代谢组学技术,研究其中酚酸类差异代谢物的相关生物学特性和代谢途径。结果表明,羊肚菌MLM和MTL子实体中共检测到102个酚酸类代谢物,共有化合物93个,羊肚菌MLM特有5个,羊肚菌MTL特有4个,羊肚菌MLM和MTL代谢物含量分别占总酚酸类的45.37%、54.63%。邻苯二甲酸二异辛酯、2-羟基-(4-羟基苯基)丙酸分别在MLM、MTL中含量最高;6’-O-阿魏酰-D-蔗糖、苯甲酰酒石酸、二氢咖啡酰葡萄糖、3,4-二甲氧基肉桂酸仅在MTL中含有,3,4-二羟基苯乙醇、丁香酸、棓因、3-O-阿魏酰奎宁酸、硫磺菊素仅在MLM含有。筛选出55个差异代谢物,共有差异代谢物46个,羊肚菌MLM特有4个,羊肚菌MTL特有5个差异代谢物,差异代谢物主要分布在新陈代谢途径、次级代谢、酪氨酸代谢等18种代谢通路中。
【Abstract】 In order to explore the differences of phenolic acid metabolites in fruitbody of Morchella esculenta, the types and contents of phenolic acids in the fruitbody of M. esculenta MLM and MTL was analyzed by utra performance liquid chromatography-tandem mass spectrometry(UPLC-MS/MS).The biological characteristics and metabolic pathways of different phenolic acids metabolites were studied by metabolomics technology. The results showed that a total of 102 phenolic acid metabolites were detected in the fruitbody of M. esculenta MLM and MTL, including 93 common compounds.The M. esculenta MLM and MTL had 5 and 4 specific metabolites, which accounting for 45.37% and 54.63% of the total phenolic acid metabolites,respectively. The contents of diisoctyl phthalate and 2-hydroxyl-(4-hydroxyphenyl) propionic acid were the highest in MLM and MTL, respectively.6’-O-feruyl-D-sucrose, benzoyltartaric acid, dihydrocaffeyl glucose and 3,4-dimethoxycinnamic acid were only found in MTL, while 3,4-dihydroxyphenylethanol, syringic acid, gallein, 3-o-feruylquinic acid and ivothrin were only found in MLM. The 55 differential metabolites were screened, including 46 differential metabolites, and M. esculenta MLM and MTL had 4 and 5 specific differential metabolites. The differential metabolites mainly distributed in 18 metabolic pathways, such as metabolic pathway, secondary metabolic pathway, and tyrosine metabolic pathway.
