抗尖孢镰刀菌贝莱斯芽孢杆菌P9培养基及发酵条件优化
Optimization of medium and fermentation conditions of Bacillus velezensis P9 resistant to Fusarium oxysporum
【摘要】 该研究以贝莱斯芽孢杆菌(Bacillus velezensis)P9为试验菌株,以其发酵液对尖孢镰刀菌(Fusarium oxysporum)的抑菌效果为考察指标,采用单因素试验及响应面法对其培养基和发酵条件进行优化。结果表明,最佳发酵培养基为:可溶性淀粉6.8 g/L、酵母浸粉18.1 g/L、MnCl20.89 g/L、FeCl20.5g/L、MgSO4·7H2O 0.5 g/L,K2HPO41.4 g/L,KH2PO40.6 g/L,pH 7.0;最佳发酵条件为:接种量4.5%,发酵温度30℃,转速240 r/min,培养时间72 h。在此优化条件下,贝莱斯芽孢杆菌P9发酵液的抑菌圈直径可达15.0 mm左右,比初始抑菌圈直径扩大1.7倍。
【Abstract】 Using Bacillus velezensis P9 as the test strain,and the antibacterial effect of its fermentation broth on Fusarium oxysporum as the investigation index,the medium formula and fermentation conditions were optimized by single factor tests and response surface methodology.The results showed that the optimal fermentation medium was as follows:soluble starch 6.8 g/L,yeast powder 18.1 g/L,Mn Cl20.89 g/L,FeCl20.5g/L,MgSO4·7H2O 0.5g/L,K2HPO41.4 g/L,KH2PO40.6 g/L,pH 7.0.The optimum fermentation conditions were as follows:inoculum 4.5%,fermentation temperature 30℃,rotation speed 240 r/min,and fermentation time 72 h.Under the optimized conditions,the diameter of the inhibition zone of B.velezensis P9 fermentation broth reached about 15.0 mm,which was 1.7 times larger than the diameter of the initial inhibition zone.
