车前子壳诱导木聚糖酶与阿拉伯糖苷酶协同制备

【摘要】 该研究以车前子壳为诱导底物,采用单因素试验和响应面试验对里氏木霉(Trichoderma reesei)诱导木聚糖酶和阿拉伯糖苷酶的协同制备条件进行优化。单因素试验结果表明,在诱导碳源为车前子壳,初始pH为5.0、底物质量浓度为20 g/L、培养时间为4.0 d、碳氮比为1.6∶1.0时,木聚糖酶和阿拉伯糖苷酶活力最高,分别为(10.59±0.33)IU/mL和(8.99±0.05)IU/mL。响应面试验结果表明,最佳产酶条件为培养时间4.0 d、底物质量浓度20 g/L、碳氮比1.6∶1.0。在此最佳条件下,木聚糖酶和阿拉伯糖苷酶活力分别为(11.42±0.15)IU/mL和(10.83±0.08)IU/mL,分别是未优化前的2.55倍和7.37倍。

【Abstract】 Using psyllium husk as induction substrate, the fermentation conditions for the synergistic preparation of xylanase and arabinosidase induced by Trichoderma reesei were optimized by single-factor tests and response surface tests. The results of single-factor tests showed that the activities of xylanase and arabinosidase were the highest under the condition of psyllium husk as induced carbon source, initial pH 5.0, substrate mass concentration 20 g/L, culture time 4.0 d, and carbon and nitrogen ratio 1.6∶1.0, which were(10.59±0.33) IU/ml and(8.99±0.05) IU/ml, respectively. The response surface tests results showed that the optimized enzyme production were as follows: culture time 4.0 d, substrate mass concentration 20 g/L, and carbon and nitrogen ratio 1.6∶1.0. Under these optimal conditions, the activities of xylanase and arabinosidase were(11.42±0.15) IU/ml and(10.83±0.08) IU/ml,which were 2.55 times and 7.37 times higher than those before optimization, respectively.
