O-GlcNAc修饰对宿主细胞抵抗类鼻疽伯克霍尔德菌感染的影响
The O-GlcNAcylation enhances the anti-infective ability of host cells against Burkholderia pseudomallei
【摘要】 目的 探讨在不同浓度葡萄糖培养下人肺癌上皮A549细胞对类鼻疽伯克霍尔德菌(Burkholderia pseudomallei,Bp)的抗感染能力的变化,探究O-GlcNAc糖基化修饰对宿主细胞抗感染能力的影响。方法 分别用含0、1、5.5、20 mmol/L葡萄糖的DMEM培养基培养A549细胞,并在此条件下用类鼻疽伯克霍尔德菌感染A549细胞,感染24 h后用Giemsa染色,显微镜下观察细胞的空斑形成面积和多核巨细胞(MNGCs)数量;CCK-8检测细胞存活率;Western blot检测O-GlcNAc糖基化修饰水平的变化。结果 A549细胞空斑形成面积、多核巨细胞数量随葡萄糖浓度的增高而减少(均P<0.01);细胞存活率随葡萄糖浓度增高呈递增趋势(P<0.01);细胞的O-GlcNAc修饰水平岁葡萄糖浓度增高呈递增趋势(P<0.05)。结论 葡萄糖有助于增强A549细胞对类鼻疽伯克霍尔德菌的抗感染能力,推测O-GlcNAc糖基化修饰在其中起调控作用,但具体机制有待进一步研究。
【Abstract】 Objective To investigate the changes of the anti-infective ability of A549 cells against Burkholderia pseudomallei(Bp) under different concentrations of glucose and to explore the effect of O-GlcNAcylation on host’s anti-infective ability. Methods A549 cells were cultured in DMEM medium containing 0,1,5.5,20 mmol/L glucose respectively, and infected with Bp under these conditions. After 24 hours of infection, the A549 cells were stained with Giemsa, the plaque area and the number of multinuclear giant cells(MNGCs) were observed under microscope. The cell viability was detected with CCK-8 kit, and O-GlcNAcylation was detected with Western blot. Results With the increase of glucose concentration, the plaque areas and the number of multinuclear giant cells of A549 cells decreased(all P<0.01),while the cell survival rate and the level of O-GlcNAcylation increased(P<0.05). Conclusion Glucose can enhance the anti-infective ability of A549 cells against Bp and speculated that O-GlcNAcylation plays an important regulatory role, but the specific mechanism should be further explored.
