濒危植物紫纹兜兰无菌萌发研究

【摘要】 以紫纹兜兰在自然状态下授粉210 d的蒴果为材料,研究不同基础培养基及添加剂对紫纹兜兰种子无菌萌发及原球茎生长的影响。结果表明,种子呈纺锤形,多数种胚发育不良;1/4 MS为种子无菌萌发的最适基础培养基;添加椰汁和香蕉汁可以明显提高种子萌发率;整体萌发生长情况最佳的培养基为1/4 MS+20 g/L蔗糖+7 g/L琼脂+100 ml/L椰汁+0.2 mg/L 6-BA+0.1 mg/L NAA,其次是1/4 MS+20 g/L蔗糖+7 g/L琼脂+100 mg/L香蕉泥+0.2mg/L 6-BA+0.1 mg/L NAA,前者萌发时间及生长速度快,但无明显增殖作用,后者萌发和生长较慢,但原球茎发生增殖分化现象明显;在萌发培养基中加少量植物激素,促使原球茎诱导分化后再进行换瓶培养能避免褐变。利用腐殖土和松树皮配比1∶1(体积比)作移栽基质,试管苗移栽成活率达90%以上。

【Abstract】 The material used in this study was the 210 days capsules produced from pollinating the self-incompatible Paphiopedilum purpuratum. The effects of different basic culture media and additives on seedling aseptic germination and protocorm growth were studied. Results showed that the seeds were spindle-shaped and most of the embryos were poorly developed. The optimal basic culture medium for aseptic seed germination was found to be 1/4 MS, while the addition of coconut milk and banana juice significantly improved the germination rate. The best overall medium for germination and growth was found to be 1/4 MS+20 g/L sucrose+7 g/L agar+100 mL/L coconut milk+0.2 mg/L 6-BA+0.1 mg/L NAA, followed by 1/4 MS+20 g/L sucrose+7 g/L agar+100 mg/L banana puree+0.2 mg/L 6-BA+0.1 mg/L NAA. The former had a faster germination time and growth rate but no significant proliferation effect, while the latter had slower germination and growth rate but obvious tuber proliferation and differentiation. Adding a small amount of plant hormones to the germination culture medium could induce protocorm differentiation before transferring to a new container, which helped to avoid browning phenomenon. Using a 1∶1 mixture of humus soil and pine bark as the cultivation substrate, the survival rate of tissue-cultured seedlings after transfer planting exceeded 90%.
