细菌靶向肿瘤治疗的研究进展

【摘要】 靶向治疗和免疫治疗的兴起与发展为治愈多种癌症带来了希望。然而,在寻求新的肿瘤治疗方法和药物过程中仍面临着安全性、特异性、可持续性和耐药性等诸多因素的持续挑战。细菌靶向肿瘤疗法的再次兴起为抗癌提供了一种独特的治疗选择,并且有望同时攻克以上这些难题。与一些其他疗法相比,肿瘤靶向细菌具有多种抗癌方式。细菌可以优先在肿瘤内定植和积累,诱导机体启动抗肿瘤免疫反应。细菌可以根据临床需求进行多样的基因编辑,从而使其靶向肿瘤并呈递特异性的抗癌制剂,实现更好的肿瘤治疗效果。本文对细菌抗肿瘤的研究成果进行归纳总结,就细菌靶向肿瘤疗法必须考量的安全性、靶向性、药物表达策略、药物传递方式和临床试验等五个方面研究进展进行了综述。

【Abstract】 The rise and development of targeted therapy and immunotherapy have brought hope to cure many kinds of cancer. However, in the process of cancer treatment exploration, there are still continuous challenges such as safety, specificity persistence and drug resistance. Bacterial targeted cancer therapy provides a unique treatment option for anti-cancer. In contrast to some other therapies, targeting bacteria has multiple ways to fight cancer. Bacteria preferentially colonize and accumulate in tumors, which can initiate anti-tumor immune responses. Bacteria can perform a variety of gene editing based on clinical needs to produce specific anticancer agents. This article summarizes the research achievements of bacterial anti-tumor therapy, and reviews the research progress in five aspects: safety, targeting, drug expression strategy, drug delivery mode and clinical trials.
