糖尿病肾脏疾病小鼠肾组织环状RNA差异表达的研究
Differential expression analysis of circular RNA in mice kidney with diabetic kidney diseas
【摘要】 目的 探讨STZ诱导的DKD小鼠环状RNA(circRNA)的差异表达。方法 8只小鼠分为正常对照组(Con)、DKD组,每组各4只。采用circRNA芯片检测各组肾组织circRNA表达谱,筛选差异表达circRNA分子,采用实时荧光定量PCR验证差异表达circRNA分子在各组肾组织中的表达变化,采用TargetScan及miRanda软件预测差异表达circRNA可能作用的miRNA靶点。结果DKD组共筛选出177个差异表达circRNA(差异倍数≥1.2且P<0.05),其中62个circRNA上调,115个circRNA下调,表达上调前5的差异circRNA是mmu_circRNA_40538、mmu_circRNA_21098、mmu_circRNA_20027、mmu_circRNA_25333和mmu_circRNA_25074,表达下调前5的差异circRNA是mmu_circRNA_19015、mmu_circRNA_41646、mmu_circRNA_011174、mmu_circRNA_29639及mmu_circRNA_28869。与Con组比较,DKD组FBG、24 h UAlb、mmu_circRNA_25074表达升高(P<0.05或P<0.01),体重、mmu_circRNA_011174及mmu_circRNA_41646表达降低(P<0.05)。结论 STZ诱导的DKD小鼠肾组织中circRNA呈差异表达,为验证circRNA在DKD中的作用提供依据。
【Abstract】 【Objective】 To explore the expression of kidney circular RNA in mice with diabetic kidney disease(DKD)induced by STZ.MethodsA total of 8 mice were averagely divided into control group and DKD group. Different expression circRNAs were detected and screened by circRNA microchip. Then they were confirmed by quantitative real-time PCR in kidney tissue and compared between control and DKD mice.Finally,miRNA that can bind to circRNA was predicted by TargetScan & miRanda.ResultsA total of177 different circRNA were found in DKD group(fold change≥1. 2 and P<0. 05). Among them,62circRNAs were upregulated and 115 circRNAs were down-regulated. The top five upregulated circRNAs were mmu_circRNA_40538, mmu_circRNA_21098,mmu_circRNA_20027,mmu_circRNA_25333 and mmu_circRNA_25074. And the top five downregulated circRNAs were mmu_circRNA_19015,mmu_circRNA_41646,mmu_circRNA_011174,mmu_circRNA_29639 and mmu_circRNA_28869. Compared with Con group,FBG,24 h UAlb,and mmu_circRNA_2507 expression were increased(P<0. 05 or P<0. 01),while weight,mmu_circRNA_011174 and mmu_circRNA_41646 expression were decreased(P<0. 05)in DKD group.ConclusionThere were different expression circRNAs in kidney of STZ-induced DKD mice. It will provide the basis for further study on the possible involvement of circRNAs in the pathogenesis of DKD.
