酸铝胁迫下红三叶种质萌发期和幼苗期生长特性及综合评价
Characterization of Growth Attributes and Comprehensive Assessment of Red Clover Germplasm at Germination and Seedling Stages under Acid-aluminum Stress
【摘要】 为评价红三叶种质耐酸铝能力强弱,以30份国内外红三叶种质材料为对象,研究其萌发期和幼苗期不同Al3+浓度胁迫下的生长特性,并根据灰色理论对红三叶耐酸铝性进行综合评价。结果表明:随着Al3+胁迫浓度的增加,红三叶相对发芽率逐渐降低,胚根变短,变粗;相对于发芽率,胚根对Al3+胁迫更为敏感,在4 mmol/L的Al3+胁迫下,胚根长度仅为对照的0.16倍。红三叶幼苗能够耐受20 mmol/L的Al3+胁迫,但在酸铝胁迫下其根系和茎叶生长受到抑制,相对根总长度、相对根表面积、相对根体积、相对根尖数、相对地上生物量、相对地下生物量、相对叶绿素含量及相对绿叶率均显著降低。不同浓度间红三叶各性状差异显著,2 mmol/L和10 mmol/L的Al3+浓度可分别作为红三叶萌发期和幼苗期耐酸铝评价的处理浓度。幼苗期的相对根表面积、相对根尖数、相对叶绿素含量和相对绿叶率可以作为红三叶耐酸铝评价的关键指标,其模型预测值与综合评价值显著相关。聚类分析可将30份红三叶种质聚类为3个不同酸铝耐受性类群,酸铝耐受性强的类群具有更高的相对根表面积、相对根尖数量、相对叶绿素含量和相对绿叶率指标值,并且各类群间关键指标性状差异显著。筛选出3个酸铝耐受性较强的红三叶种质EM00232、EM00222和CF022235,可作为耐酸铝红三叶新品种选育的亲本材料。
【Abstract】 This study aimed to evaluate the acid-aluminum (Al3+) tolerance in 30 red clover germplasm lines during the stages of germination and seedling growth,employing a grey theory model.The results showed a gradual reduction in relative germination rate that as the concentration of Al3+increased,concur‐rently leading to shorter and thicker radicles.The radicle exhibited greater sensitivity to Al3+stress,its length diminishing to a mere 0.16 of the control’s length at an Al3+concentration of 4 mmol/L.Red clover seedlings demonstrated tolerance to Al3+stress up to 20 mmol/L.However,at higher concentrations,there was a significant reduction in relative total root length,relative root surface area,relative root vol‐ume,relative root tip number,relative aboveground biomass,relative underground biomass,relative chlo‐rophyll content,and relative green leaf ratio,in comparison to the control.Notable differences in the major traits of red clover were observed across various Al3+concentrations.Evaluating the Al3+tolerance of dif‐ferent red clover germplasm materials was performed at germination and seedling stages,at optimal Al3+concentrations of 2 mmol/L and 10 mmol/L,respectively.A prediction model was established to estimate the Al3+tolerance for each red clover germplasm line,based on four key factors:relative root surface area,relative root tip number,relative chlorophyll content,and relative green leaf ratio,showing a significant correlation with the multivariate evaluation value.Through clustering analysis,30 red clover germplasm resources were categorized into three distinct Al3+tolerant groups.The groups exhibiting higher Al3+toler‐ance displayed increased values in relative root surface area,relative root tip number,relative chlorophyll content,and relative green leaf rate,presenting significantly differences among groups.Notably,three red clover germplasms,namely EM00232,EM00222 and CF022235,displayed high Al3+tolerance,suggest‐ing their potential as parent materials for breeding Al3+-tolerant varieties of red clover.
