基于HPLC指纹图谱成分含量和化学模式识别不同产地防风与水防风
Identification of Saposhnikovia divaricata and Libanotis laticalycina from different producing areas based on component content in HPLC fingerprint and chemical pattern recognition
【摘要】 目的 基于特征图谱及主要成分的含量,并结合化学模式,比较防风与水防风的化学成分差异。方法 Phenomenex Kinetex?XB-C18色谱柱(250 mm×4.6 mm, 5μm);流动相乙腈-水,梯度洗脱;体积流量1.0 mL/min;柱温30℃;检测波长254 nm。采用“中药色谱指纹图谱相似度评价系统(2012A)”进行相似度评价,建立共有模式。以色谱峰信息为数据来源,采用3种化学模式识别方法综合分析防风、水防风药材差异。结果 建立了不同产地防风的HPLC指纹图谱共有模式,标定了17个特征峰,相似度基本>0.9。聚类分析和主成分分析能对不同产地防风及伪品水防风进行明显区分;正交偏最小二乘法判别分析表明导致差异的主要化学标记物为升麻素苷、升麻素、6″-O-apiosyl-5-O-methylvisammiosid、亥茅酚苷、补骨脂素、花椒毒素、3-O-乙酰基亥茅酚、异欧前胡素。5-O-甲基维斯阿米醇苷与亥茅酚苷含量之和与升麻素含量的比值在防风与水防风中区别较大,具有一定的鉴别意义。结论 该方法可用于分析防风、水防风药材质量的差异性,为规范防风药材使用的评价提供参考。
【Abstract】 AIM To identify the similarities and differences between Saposhnikovia divaricata and Libanotis laticalycina based on the characteristic map, the contents of main components, and the chemical model.METHODS The determination was performed on the 30 ℃ thermostatic Phenomenex Kinetex? XB-C18 column(250 mm×4.6 mm, 5 μm), with acetonitrile-water as the mobile phase, the gradient elution flowing at the rate of 1.0 mL/min, and the detective wavelength working at 254 nm. The similarity evaluation was carried out by using the “Similarity Evaluation System of Chinese Medicine Chromatographic Fingerprint(2012A)” for the establishment of the common model as well. Given the chromatographic peak information, the comprehensive analysis of the data of S. divaricata and L. laticalycina were conducted using three chemical pattern recognition method.RESULTS In the established common HPLC fingerprint pattern, seventeen characteristic peaks were calibrated, sharing more than 0.9 similarity. The medicinal materials from different origins were clearly distinguished by cluster analysis and principal component analysis. According to the result of discriminant analysis by orthogonal partial least squares method, cimicifuga glycoside, cimicifugain, 6″-O-apiosyl-5-O-methylvisammiosid, haim oside, psoralen, xanthotoxin, 3-O-acetylheliofol, isoimperatorin were the main chemical markers leading to the difference. The very difference in content ratio of 5-O-methylvisamimitol glycoside and haim oside to cimicifugain in S. divaricata and L. laticalycina was of significant value in their identification.CONCLUSION Our method can effectively identify the quality difference of S. divaricata and L. laticalycina, providing an evaluation reference for the standard use of S. divaricate.
