香砂六君子汤对脾胃虚弱型功能性消化不良大鼠胃动力的作用
Effects of Xiangsha Liujunzi Decoction on gastric motility of rats with functional dyspepsia of Spleen-Stomach Deficiency Pattern
【摘要】 目的 探讨香砂六君子汤对脾胃虚弱型功能性消化不良大鼠胃动力的作用。方法 将大鼠随机分为空白组、模型组、阳性药组及香砂六君子汤高、中、低剂量组,采用碘乙酰胺+饥饱饮食+游泳力竭方法复制脾胃虚弱型功能性消化不良大鼠模型,给予相应药物干预14 d。观察各组大鼠一般情况,测定各组大鼠体质量、3 h进食量、胃排空率,HE染色观察大鼠胃窦组织病理学改变,ELISA、RT-qPCR、Western blot法检测大鼠胃窦组织AMPK、TSC2、mTOR、Ghrelin表达。结果 与空白组比较,模型组大鼠体质量、3 h进食量、胃排空率和胃窦组织AMPK、TSC2、Ghrelin mRNA和蛋白表达均降低(P<0.05),mTOR mRNA和蛋白表达均升高(P<0.05);与模型组比较,香砂六君子汤高、中剂量组大鼠体质量、3 h进食量、胃排空率和胃窦组织AMPK、TSC2、Ghrelin mRNA和蛋白表达均升高(P<0.05),mTOR mRNA和蛋白表达均降低(P<0.05)。结论 香砂六君子汤能改善脾胃虚弱型功能性消化不良大鼠胃动力障碍,其机制与激活AMPK/TSC2/mTOR信号通路,促进Ghrelin表达相关。
【Abstract】 AIM To investigate the effects of Xiangsha Liujunzi Decoction on gastric motility of rats with functional dyspepsia due to Spleen-Stomach Deficiency.METHODS The rats were randomly divided into the blank group, the model group, the positive drug group and high, medium and low dose Xiangsha Liujunzi Decoction groups. The rat models of functional dyspepsia of Spleen-Stomach Deficiency Pattern were induced by the combinative use of iodoacetamide, starvation and refeeding, and swimming exhaustion, followed by the 14 days corresponding drugs administration. Subsequently, the rats had their general physical conditions observed; their body weight, 3-hour food intake and gastric emptying rate determined; their pathological changes of gastric antrum observed by HE staining; and their expressions of AMPK, TSC2, mTOR and Ghrelin in gastric antrum detected by ELISA, RT-qPCR and Western blot.RESULTS Compared with the blank group, the model group displayed decreased body weight, 3-hour food intake, gastric emptying rate, and the mRNA and protein expressions of AMPK, TSC2 and Ghrelin in gastric antrum(P<0.05), and increased mRNA and protein expression of mTOR(P<0.05). Compared with the model group, high and medium dose Xiangsha Liujunzi Decoction groups demonstrated increased body weight, 3-hour food intake, gastric emptying rate, and mRNA and protein expressions of AMPK, TSC2 and Ghrelin(P<0.05), and decreased mRNA and protein expression of mTOR(P<0.05).CONCLUSION Xiangsha Liujunzi Decoction may improve the gastric motility disorder in rats with functional dyspepsia of Spleen-Stomach Deficiency Pattern via activating AMPK/TSC2/mTOR signaling pathway and promoting Ghrelin expression.
