UPLC-MS/MS法同时测定连花清瘟胶囊中11种成分
Simultaneous determination of eleven constituents in Lianhua Qingwen Capsules by UPLC-MS/MS
【摘要】 目的 建立UPLC-MS/MS法同时测定连花清瘟胶囊中红景天苷、绿原酸、连翘酯苷A、连翘苷、槲皮素、木犀草素、山柰酚、大黄酸、甘草酸、大黄素、大黄素甲醚的含量。方法 该药物75%甲醇提取液的分析采用XB-C18色谱柱(100 mm×3 mm, 2.6μm);流动相乙腈-甲醇-水(含10 mmo/L乙酸铵和0.1%甲酸),梯度洗脱;体积流量0.5 mL/min;柱温40℃;电喷雾离子源;正负离子检测;多反应监测模式。结果 11种成分在各自范围内线性关系良好(r≥0.994 6),平均加样回收率85.5%~110.3%,RSD 1.2%~3.9%。结论 该方法准确快速,重复性好,可用于连花清瘟胶囊的质量控制。
【Abstract】 AIM To establish a UPLC-MS/MS method for the simultaneous content determination of salidroside, chlorogenic acid, forsythiaside A, forsythin, quercetin, luteolin, kaempferol, rhein, glycyrrhizic acid, emodin and emodin-3-methyl ether in Lianhua Qingwen Capsules.METHODS The analysis of 75%methanol extract of this drug was performed on a 40 ℃ thermostatic XB-C18 column(100 mm×3 mm, 2.6 μm), with the mobile phase comprising of acetonitrile-methanol-water(containing 10 mmol/L ammonium acetate and 0.1%formic acid) flowing at 0.5 mL/min in a gradient elution manner, and electron spray ionization source was adopted in positive and negative ion detection with multiple reaction monitoring mode.RESULTS Eleven constituents showed good linear relationships within their own ranges(r≥0.994 6), whose average recoveries were 85.5%-110.3%with the RSDs of 1.2%~3.9%.CONCLUSION This accurate, rapid and reproducible method can be used for the quality control of Lianhua Qingwen Capsules.
