STIM1和STIM2在人循环纤维细胞中的表达模式分析及鉴定
Analysis and Identification of Expression Patterns of STIM1 and STIM2 in Human Circulating Fibroblasts
【摘要】 目的:探讨SOC通道的相关基因STIM1和STIM2在人循环纤维细胞中的表达模式鉴定及功能。方法:比较STIM1和STIM2在人各部位的表达模式。采集人外周静脉血,分离出单个核细胞,体外培养分化为循环纤维细胞。采用RT-PCR以及western blotting检测循环纤维细胞中STIM1和STIM2的表达情况。并检测SOC通道促进剂IL-4和抑制剂SKF-96365对循环纤维细胞分化的影响及对STIM1和STIM2表达的影响。结果:STIM1和STIM2在人体组织组成型表达,且STIM1的表达量均高于STIM2。RT-PCR和western blotting检测显示循环纤维细胞表达STIM1和STIM2,并且SOC通道促进剂IL-4对循环纤维细胞分化成熟具有促进作用,SOC通道抑制剂SKF96365对循环纤维细胞分化成熟具有抑制作用。结论:人循环纤维细胞中表达STIM1和STIM2,其中STIM1可能起主导作用,且人循环纤维细胞中STIM1和STIM2的mRNA和蛋白表达水平受IL-4和SKF的影响。
【Abstract】 Objective To investigate the expression patterns and function of SOC channel related genes STIM1 and STIM2 in circulating fibroblasts. Methods Compare the expression patterns of STIM1 and STIM2 in various parts of the human body, collect human peripheral venous blood, isolate mononuclear cells, and cultivate them in vitro to differentiate into circulating fibroblasts. RT-PCR and Western blotting were used to detect the expression of STIM1 and STIM2 in circulating fibroblasts. The effects of SOC channel promoter IL-4 and inhibitor SKF-96365 on the differentiation of circulating fibroblasts and the expression of STIM1 and STIM2 were also detected. Results STIM1 and STIM2 were expressed in all human tissues, and the expression of STIM1 was higher than that of STIM2. The detection of RT-PCR and western blotting showed that circulating fibroblasts expressed STIM1 and STIM2. SOC channel promoter IL-4 could promote the differentiation and maturation of circulating fibroblasts, and SOC channel inhibitor SKF96365 could inhibit the differentiation and maturation of circulating fibroblasts. Conclusions STIM1 and STIM2 are expressed in human circulating fibroblasts, with STIM1 possibly playing a dominant role. The mRNA and protein expression levels of STIM1 and STIM2 in human circulating fibroblasts are influenced by IL-4 and SKF.
