人参多糖对肠上皮屏障的增强作用及其机制
Enhancing Effect and Mechanism of Panax ginseng Polysaccharide on Intestinal Epithelial Barriers
【摘要】 目的 观察人参多糖对大鼠小肠上皮细胞(IEC-6)渗透性、黏附连接蛋白表达及细胞钙离子(Ca2+)水平的影响,探讨益气健脾中药人参肠黏膜保护作用机制。方法 设置不同实验条件[正常含钙培养、含钙培养+二氟甲基鸟氨酸(DFMO)、无钙培养]以观察人参多糖的作用;Transwell小室酚红透过法检测细胞间渗透性;Western blotting法检测黏附连接蛋白(E-cadherin、α-catenin和β-catenin)表达;激光共聚焦显微镜观察细胞Ca2+水平。结果 (1)正常含钙培养时,人参多糖(40、80、160 mg·L-1,下同)能降低细胞间渗透性、提高E-cadherin、α-catenin和β-catenin表达及细胞Ca2+水平(P<0.05或P<0.01);(2)含钙培养+DFMO负荷时,细胞间渗透性增加、E-cadherin和α-catenin表达及Ca2+水平降低(P<0.05或P<0.01),人参多糖可改善DFMO所致的细胞间渗透性增加、E-cadherin和α-catenin表达及Ca2+水平降低(P<0.05或P<0.01);(3)无钙培养可致细胞间渗透性增加及E-cadherin、α-catenin和β-catenin表达降低(P<0.05或P<0.01),人参多糖可改善无钙培养所致的细胞间渗透性增加及E-cadherin和α-catenin表达下降(P<0.05或P<0.01)。结论 人参多糖具有增强肠上皮屏障的作用,其机制与提高细胞Ca2+水平及黏附连接蛋白表达有关。
【Abstract】 Objective To investigate the potential effect of Panax ginseng polysaccharide on the permeability, the expression of adhesion junction proteins, and calcium ion level in rat small intestinal epithelial cells(IEC-6),and to explore the mechanism of Chinese herbs with the actions of strengthening qi and invigorating spleen in intestinal mucosal protection. Methods Different experimental conditions [normal Ca2+-containing culture, Ca2+-containing +DFMO(difluoromethylornithine, polyamine synthesis inhibitors) culture, and Ca2+-free culture] were set to investigate the effect of Panax ginseng polysaccharide; the transwell cell phenol red permeability was measured to observe the intercellular permeability; Western blotting was used to determine the expression of adhesion junction proteins(E-cadherin, α-catenin, and β-catenin);laser confocal microscopy was used to observe the level of intracellular calcium. Results(1)When cultured in normal Ca2+-containing medium, the result showed that Panax ginseng polysaccharide(40,80 or 160 mg·L-1,below) can reduce intercellular permeability, improve the expression of adhesion junction proteins(E-cadherin, α-catenin and β-catenin) and the level of intracellular calcium(P<0.05 or P<0.01);(2)In Ca2+-containing +DFMO culture, the intercellular permeability increased and the expression of adhesion junction proteins(E-cadherin, α-catenin)and the level of intracellular calcium decreased(P<0.05 or P<0.01),Panax ginseng polysaccharide could ameliorate the increase of intercellular permeability and the reduction of adhesion junction proteins(E-cadherin, α-catenin)and the level of intracellular calcium induced by DFMO(P<0.05 or P<0.01);(3)In Ca2+-free culture, the intercellular permeability increased and the expression of adhesion junction proteins(E-cadherin、α-catenin and β-catenin)and the level of intracellular calcium decreased(P<0.05),Panax ginseng polysaccharide could ameliorate the increase of intercellular permeability and decrease of adhesion junction proteins(E-cadherin, α-catenin and β-catenin, P<0.05 or P<0.01 compared with the model group). Conclusion Panax ginseng polysaccharide can enhance the intestinal epithelial barrier, and the mechanism is related to its improvement on the level of intracellular calcium and the expression of adhesion junction proteins.
