姜黄素和栓菌酸联用促吸收增效作用研究
Study on absorption promotion and synergistic effect of curcumin and trametenolic acid B
【摘要】 目的:观察姜黄素(CUR)和栓菌酸(TAB)联用,对吸收和抗肿瘤活性的影响,为提高CUR、TAB生物利用度提供依据。方法:采用MTT法研究CUR和TAB联用对SGC7901细胞增殖的影响;采用高效液相色谱法测定SGC7901细胞内CUR、TAB的积累情况及相互影响;采用高效液相色谱法测定CUR和TAB联用对大鼠血药浓度的影响。结果:MTT结果显示CUR(12μg·mL-1)与TAB(40μg·mL-1)联用对SGC7901细胞增殖有明显的抑制作用,联合用药指数<1,说明CUR与TAB联用呈现协同作用;CUR(24μg·mL-1)与TAB(80μg·mL-1)联用后,细胞内CUR浓度是单用的2.98倍,TAB浓度是单用的1.63倍,结果显示CUR和TAB在体外有相互促吸收作用;大鼠灌胃CUR后Cmax是6.48μg·mL-1,CUR和TAB联用组Cmax为14.74μg·mL-1;大鼠灌胃TAB后Cmax是16.77μg·mL-1,联用后TAB的Cmax是48.67μg·mL-1,以上结果显示在体内实验中CUR和TAB能相互促进吸收。结论:CUR与TAB联用具相互促进吸收,且协同抑制SGC7901细胞增殖的双重作用。
【Abstract】 Objective:To provide evidence for improving the bioavailability of curcumin(CUR) and trametenolic acid B(TAB) by observing the effect of CUR and TAB on absorption and anti-tumor activity. Methods: The growth of SGC7901 cells treated with CUR and TAB was evaluated by MTT assays. The accumulation of CUR and TAB in the SGC7901 cells was analyzed by high performance liquid chromatogram(HPLC). The effect of CUR combined with TAB on blood drug concentration in rats was determined by HPLC. Results: CUR(12 μg·mL-1) combined with TAB(40 μg·mL-1) inhibited the proliferation of SGC7901 cells, and the drug combination index <1, indicating that CUR and TAB had a synergistic effect. After CUR(24 μg·mL-1) was combined with TAB(80 μg·mL-1), the concentration of CUR in cells was 2.98 times of CUR alone, the concentration of TAB was 1.63 times of TAB alone. These results showed that CUR and TAB promoted the absorption of each other in vitro. After oral administration, the Cmax of CUR in rats was 6.48 μg·mL-1, and Cmax of CUR of the combined group was 14.74 μg·mL-1. The Cmax of TAB in rats was 16.77 μg·mL-1 after oral administration and and 48.67 μg·mL-1 after combined with TAB. Conclusion: The combination of CUR and TAB can promote absorption mutually, and synergistically inhibit the proliferation of SGC7901 cells.
