化痰祛湿方对自发性高血压大鼠的肾脏保护作用

【摘要】 目的 探讨化痰祛湿方对自发性高血压大鼠(SHR)的肾脏保护作用。方法 40只SHR随机分为四组,每组10只。A、B组分别采用化痰祛湿方4、2 g/kg灌胃,C组马来酸依那普利0.02 g/kg灌胃,D组等量生理盐水0.2 mL/kg灌胃。10只京都种Wistar大鼠为空白对照(E)组,同D组灌胃。五组每日给药1次,连续给药8周。检测五组大鼠血清肌酐、血尿素氮、血管紧张素Ⅱ和一氧化氮水平,HE和Masson染色观察肾脏病理变化和肾脏纤维化程度,Western blot法检测磷脂酰肌醇3激酶(PI3K)、Akt和哺乳动物雷帕霉素受体蛋白(mTOR)蛋白表达。结果 成功建立SHR模型。与D组比较,A、C组大鼠血清肌酐、血尿素氮、血管紧张素Ⅱ水平下降,而一氧化氮水平升高(P<0.05或P<0.01)。HE染色和Masson染色结果示,A、C组肾脏病理变化改善较D组更明显。与E组比较,D组PI3K、Akt和mTOR蛋白表达升高;与D组比较,A、C组PI3K、Akt、mTOR蛋白表达降低,B组Akt和mTOR蛋白表达降低(P<0.05)。结论 化痰祛湿方可能通过抑制PI3K/Akt信号通路,从而发挥对SHR肾脏的保护作用。

【Abstract】 Objective To investigate the protective effect of Huatan Qushi decoction on kidney in spontaneous hypertension rats(SHR).Methods Forty of SHR were randomly divided into four groups with 10 rats each.Groups of A, B, C and D were with intragastric administration of Huatan Qushi decoction 4 g/kg and 2 g/kg, enalapril maleate 0.02 g/kg, and normal saline 0.2 mL/kg, respectively. Ten Wistar-Kyoto rats were set as blank controls(group E),which were administered as the same as group D. Five groups were given once per day for 8 weeks.Serum creatinine, blood urea nitrogen, angiotensin Ⅱ and nitric oxide were detected, and renal pathological changes and renal fibrosis degree were observed by HE and Masson staining in five groups. The expressions of phosphatidylinositol 3 kinase(PI3K), Akt and mammalian target of rapamycin(mTOR) were detected by Western blot.Results The rat models with hypertension were established successfully. Compared with group D, the levels of serum creatinine, blood urea nitrogen and angiotensin Ⅱ in groups of A and C were decreased, while nitric oxide was increased(P<0.05 or P<0.01). The results of HE staining and Masson staining showed that the improvement of renal pathological changes was more obvious in groups of A and C. Compared with group E, the protein expressions of PI3K, Akt and mTOR were increased in group D(P<0.05). Compared with group D, the protein expressions of PI3K, Akt and mTOR were decreased in groups of A and C, while the protein expressions of Akt and mTOR were decreased in group B(P<0.05).Conclusion Huatan Qushi decoction may play a protective role in the kidney of SHR by inhibiting PI3K/Akt signaling pathway.
