低强度电刺激对神经损伤性ED大鼠勃起功能影响

【摘要】 目的 评估低强度电刺激(LIES)对神经损伤性勃起功能障碍(ED)大鼠勃起功能(EF)影响。方法 雄性SD大鼠随机分为3组:Sham组、双侧海绵体神经损伤(BCNI)组和LIES+BCNI组,每组10只大鼠。LIES治疗7 d后,通过对比大鼠最大海绵体内压/平均动脉压(ICPmax/MAP)评估阴茎EF变化;通过ELISA试剂盒测量海绵体内活性氧(ROS)蛋白含量;通过Western blot方法检查海绵体中转化生长因子-β1 (TGF-β1)、白细胞介素-6 (IL-6)和C反应蛋白(CRP)蛋白表达;通过免疫荧光染色评估海绵体中α-平滑肌肌动蛋白(α-SMA)蛋白表达。结果 与Sham组相比,BCNI组大鼠ICPmax/MAP降低(P<0.05);与BCNI组比,BCNI+LIES组大鼠ICPmax/MAP升高(P<0.05)。与Sham组相比,BCNI组大鼠海绵体ROS含量增高(P<0.05);与BCNI组比,BCNI+LIES组大鼠海绵体ROS含量降低(P<0.05)。与Sham组相比,BCNI组大鼠海绵体TGF-β1、IL-6和CRP蛋白表达增加(P<0.05);与BCNI组比,BCNI+LIES组大鼠海绵体TGF-β1、IL-6和CRP蛋白表达降低(P<0.05)。与Sham组相比,BCNI组海绵体内α-SMA蛋白表达降低(P<0.05);与BCNI组比,BCNI+LIES组大鼠海绵体α-SMA蛋白表达增加(P<0.05)。结论 LIES治疗不仅改善BCNI后大鼠EF,而且对海绵体组织具有保护作用,包括防止氧化应激、炎症和平滑肌细胞丢失。

【Abstract】 【Objective】 To evaluate the effect of low intensity electrical stimulation(LIES) on erectile function(EF) in rats with nerve injury induced erectile dysfunction(ED). 【Methods】 Male SD rats were randomly divided into 3 groups: Sham group, bilateral cavernous nerve injury(BCNI) group and LIES+BCNI group, with 10 rats in each group. After 7 days of treatment with LIES, the changes of penile EF were evaluated by comparing the maximum intracavernous pressure/mean arterial pressure(ICPmax/MAP) of rats. The content of reactive oxygen species(ROS) protein in cavernous body was measured by ELISA kit. The protein expression of transforming growth factor-β1(TGF-β1), interleukin-6(IL-6) and C-reactive protein(CRP) in cavernous body was examined by Western blot. Immunofluorescence staining was used to evaluate the α-smooth muscle actin(α-SMA) protein expression. 【Results】 Compared with Sham group, ICPmax/MAP of rats in BCNI group decreased(P<0.05); compared with BCNI group, ICPmax/MAP in BCNI+LIES group increased(P<0.05). Compared with Sham group, the content of ROS in cavernous body of rats in BCNI group increased(P<0.05); compared with BCNI group, the content of ROS in cavernous body of rats in BCNI+LIES group decreased(P<0.05). Compared with Sham group, the expression of TGF-β1, IL-6 and CRP protein in cavernous body of rats in BCNI group increased(all P<0.05); compared with BCNI group, the expression of TGF-β1, IL-6 and CRP protein in cavernous body of rats in BCNI+LIES group decreased(all P<0.05). Compared with Sham group, the expression of α-SMA protein in BCNI group was decreased(P<0.05); compared with BCNI group, the α-SMA protein expression in cavernous body of BCNI+LIES group was increased(P<0.05). 【Conclusion】 LIES treatment not only improved EF in rats after BCNI, but also had protective effects on cavernous body, including prevention of oxidative stress, inflammation, and smooth muscle cell loss.
